Abstract

Alzheimer's disease (AD) is a progressive degenerative disorder characterized by memory loss and cognitive decline. However, there are limited studies on preventing or stopping the advanced neuronal loss that occurs during or before the disease. Our project aims to provide a prophylactic application against AD and similar neurodegenerative diseases. Exosomes are small particles that can cross biological membranes like the blood-brain barrier. They are being used as drug carrier systems and have attracted attention recently. In AD and similar diseases, exposure to oxidant molecules increases intracellular reactive oxygen species, which leads to incorrect protein folding and plays a role in disease pathogenesis. Alpha-tocopherol is a natural antioxidant compound that is therapeutically effective in preventing protein aggregation by stopping the increase in intracellular reactive oxygen levels. This study cultured glioblastoma (U-87) cells under appropriate conditions and exosome isolation was performed from the 8th passage. Experiments were carried out to determine the zeta potential and total protein amount, measure drug loading capacity, and take TEM images to characterize the isolated exosomes. Preliminary experiments are shared in this study.
