Effects of atorvastatin-loaded PEGylated liposome on focused magnetic stimulation in post-stroke

Abstract

We investigated whether a synergistic effect would emerge in post-stroke treatment by opening the blood-brain barrier (BBB) with focused magnetic stimulation (MagStim) and delivering Atorvastatin-Loaded PEGylated Liposomes (LipoStatin). Two weeks after middle cerebral artery occlusion (MCAO), magnetic nanoparticles (MNP) were injected at a concentration of 15mg/ml, followed by 30 minutes of MagStim. The procedure was conducted over a week with MagStim, and magnetic nanoparticles (MNP) were injected three times at two-day intervals. After MagStim, LipoStatin at 10mg/kg was administered intravenously for a week. According to the treatment, we investigated motor function, neuro-inflammation, blood-brain-barrier (BBB) integrity, and neurogenesis at three weeks after stroke. (Sham vs Control vs LipoStatin vs MagStim+LipoStatin). The MagStim+LipoStatin group showed improved motor function compared to the Control and LipoStatin groups. In addition, the expression of inflammatory markers such as IL-1β, IL-6, TNF-α, MCP-1and Iba1 were significantly reduced, and neurorestoration factors were increased dramatically in the MagStim+LipoStatin group compared to the Control and LipoStatin group in the PCR. Significant improvement was also observed in protein expression for BBB integrity and immunofluorescence for neurogenesis in the MagStim+LipoStatin group. In conclusion, our study demonstrates the beneficial synergistic effects of MagStim for blood-brain barrier opening and delivery of LipoStatin in subacute ischemic stroke. These findings provide novel evidence of the beneficial therapeutic effects of nanoparticles and underscore the need for future advancements in promising non-invasive MagStim methods for drug delivery in post-stroke treatment.
