Perceptions of responsiveness and rejection in romantic relationships. What are the implications for individuals and relationship functioning?

From birth, human beings need to be embedded into social ties to function best, because other individuals can provide us with a sense of belonging, which is a fundamental human need. One of the closest bonds we build throughout our life is with our intimate partners. When the relationship involves intimacy and when both partners accept and support each other’s needs and goals (through perceived responsiveness) individuals experience an increase in relationship satisfaction as well as physical and mental well-being. However, feeling rejected by a partner may impair the feeling of connectedness and belonging, and affect emotional and behavioural responses. When we perceive our partner to be responsive to our needs or desires, in turn we naturally strive to respond positively and adequately to our partner’s needs and desires. This implies that individuals are interdependent, and changes in one partner prompt changes in the other. Evidence suggests that partners regulate themselves and co-regulate each other in their emotional, psychological, and physiological responses. However, such processes may threaten the relationship when partners face stressful situations or interactions, like the transition to parenthood or rejection. Therefore, in this presentation, I will provide evidence for the role of perceptions of being accepted or rejected by a significant other on individual and relationship functioning, while considering the contextual settings. The three studies presented here explore romantic relationships, and how perceptions of rejection and responsiveness from the partner impact both individuals, their physiological and their emotional responses, as well as their relationship dynamics.

How curiosity affects learning and information seeking via the dopaminergic circuit

Over the last decade, research on curiosity – the desire to seek new information – has been rapidly growing. Several studies have shown that curiosity elicits activity within the dopaminergic circuit and thereby enhances hippocampus-dependent learning. However, given this new field of research, we do not have a good understanding yet of (i) how curiosity-based learning changes across the lifespan, (ii) why some people show better learning improvements due to curiosity than others, and (iii) whether lab-based research on curiosity translates to how curiosity affects information seeking in real life. In this talk, I will present a series of behavioural and neuroimaging studies that address these three questions about curiosity. First, I will present findings on how curiosity and interest affect learning differently in childhood and adolescence. Second, I will show data on how inter-individual differences in the magnitude of curiosity-based learning depend on the strength of resting-state functional connectivity within the cortico-mesolimbic dopaminergic circuit. Third, I will present findings on how the level of resting-state functional connectivity within this circuit is also associated with the frequency of real-life information seeking (i.e., about Covid-19-related news). Together, our findings help to refine our recently proposed framework – the Prediction, Appraisal, Curiosity, and Exploration (PACE) framework – that attempts to integrate theoretical ideas on the neurocognitive mechanisms of how curiosity is elicited, and how curiosity enhances learning and information seeking. Furthermore, our findings highlight the importance of curiosity research to better understand how curiosity can be harnessed to improve learning and information seeking in real life.

Assigning credit through the "other” connectome

Learning in neural networks requires assigning the right values to thousands to trillions or more of individual connections, so that the network as a whole produces the desired behavior. Neuroscientists have gained insights into this “credit assignment” problem through decades of experimental, modeling, and theoretical studies. This has suggested key roles for synaptic eligibility traces and top-down feedback signals, among other factors. Here we study the potential contribution of another type of signaling that is being revealed in greater and greater fidelity by ongoing molecular and genomics studies. This is the set of modulatory pathways local to a given circuit, which form an intriguing second type of connectome overlayed on top of synaptic connectivity. We will share ongoing modeling and theoretical work that explores the possible roles of this local modulatory connectome in network learning.

Is Theory of Mind Analogical? Evidence from the Analogical Theory of Mind cognitive model

Theory of mind, which consists of reasoning about the knowledge, belief, desire, and similar mental states of others, is a key component of social reasoning and social interaction. While it has been studied by cognitive scientists for decades, none of the prevailing theories of the processes that underlie theory of mind reasoning and development explain the breadth of experimental findings. I propose that this is because theory of mind is, like much of human reasoning, inherently analogical. In this talk, I will discuss several theory of mind findings from the psychology literature, the challenges they pose for our understanding of theory of mind, and bring in evidence from the Analogical Theory of Mind (AToM) cognitive model that demonstrates how these findings fit into an analogical understanding of theory of mind reasoning.

Magnetic Handshake Materials

Biological materials gain complexity from the programmable nature of their components. To manufacture materials with comparable complexity synthetically, we need to create building blocks with low crosstalk so that they only bind to their desired partners. Canonically, these building blocks are made using DNA strands or proteins to achieve specificity. Here we propose a new materials platform, termed Magnetic Handshake Materials, in which we program interactions through designing magnetic dipole patterns. This is a completely synthetic platform, enabled by magnetic printing technology, which is easier to both model theoretically and control experimentally. In this seminar, I will give an overview of the development of the Magnetic Handshake Materials platform, ranging from interaction, assembly to function design.

Credit Assignment in Neural Networks through Deep Feedback Control

The success of deep learning sparked interest in whether the brain learns by using similar techniques for assigning credit to each synaptic weight for its contribution to the network output. However, the majority of current attempts at biologically-plausible learning methods are either non-local in time, require highly specific connectivity motives, or have no clear link to any known mathematical optimization method. Here, we introduce Deep Feedback Control (DFC), a new learning method that uses a feedback controller to drive a deep neural network to match a desired output target and whose control signal can be used for credit assignment. The resulting learning rule is fully local in space and time and approximates Gauss-Newton optimization for a wide range of feedback connectivity patterns. To further underline its biological plausibility, we relate DFC to a multi-compartment model of cortical pyramidal neurons with a local voltage-dependent synaptic plasticity rule, consistent with recent theories of dendritic processing. By combining dynamical system theory with mathematical optimization theory, we provide a strong theoretical foundation for DFC that we corroborate with detailed results on toy experiments and standard computer-vision benchmarks.

Using extra-hippocampal cognitive maps for goal-directed spatial navigation

Goal-directed navigation requires precise estimates of spatial relationships between current position and future goal, as well as planning of an associated route or action. While neurons in the hippocampal formation can represent the animal’s position and nearby trajectories, their role in determining the animal’s destination or action has been questioned. We thus hypothesize that brain regions outside the hippocampal formation may play complementary roles in navigation, particularly for guiding goal-directed behaviours based on the brain’s internal cognitive map. In this seminar, I will first describe a subpopulation of neurons in the retrosplenial cortex (RSC) that increase their firing when the animal approaches environmental boundaries, such as walls or edges. This boundary coding is independent of direct visual or tactile sensation but instead depends on inputs from the medial entorhinal cortex (MEC) that contains spatial tuning cells, such as grid cells or border cells. However, unlike MEC border cells, we found that RSC border cells encode environmental boundaries in a self-centred egocentric coordinate frame, which may allow an animal for efficient avoidance from approaching walls or edges during navigation. I will then discuss whether the brain can possess a precise estimate of remote target location during active environmental exploration. Such a spatial code has not been described in the hippocampal formation. However, we found that neurons in the rat orbitofrontal cortex (OFC) form spatial representations that persistently point to the animal’s subsequent goal destination throughout navigation. This destination coding emerges before navigation onset without direct sensory access to a distal goal, and are maintained via destination-specific neural ensemble dynamics. These findings together suggest key roles for extra-hippocampal regions in spatial navigation, enabling animals to choose appropriate actions toward a desired destination by avoiding possible dangers.

Capacitance clamp - artificial capacitance in biological neurons via dynamic clamp

A basic time scale in neural dynamics from single cells to the network level is the membrane time constant - set by a neuron’s input resistance and its capacitance. Interestingly, the membrane capacitance appears to be more dynamic than previously assumed with implications for neural function and pathology. Indeed, altered membrane capacitance has been observed in reaction to physiological changes like neural swelling, but also in ageing and Alzheimer's disease. Importantly, according to theory, even small changes of the capacitance can affect neuronal signal processing, e.g. increase network synchronization or facilitate transmission of high frequencies. In experiment, robust methods to modify the capacitance of a neuron have been missing. Here, we present the capacitance clamp - an electrophysiological method for capacitance control based on an unconventional application of the dynamic clamp. In its original form, dynamic clamp mimics additional synaptic or ionic conductances by injecting their respective currents. Whereas a conductance directly governs a current, the membrane capacitance determines how fast the voltage responds to a current. Accordingly, capacitance clamp mimics an altered capacitance by injecting a dynamic current that slows down or speeds up the voltage response (Fig 1 A). For the required dynamic current, the experimenter only has to specify the original cell and the desired target capacitance. In particular, capacitance clamp requires no detailed model of present conductances and thus can be applied in every excitable cell. To validate the capacitance clamp, we performed numerical simulations of the protocol and applied it to modify the capacitance of cultured neurons. First, we simulated capacitance clamp in conductance based neuron models and analysed impedance and firing frequency to verify the altered capacitance. Second, in dentate gyrus granule cells from rats, we could reliably control the capacitance in a range of 75 to 200% of the original capacitance and observed pronounced changes in the shape of the action potentials: increasing the capacitance reduced after-hyperpolarization amplitudes and slowed down repolarization. To conclude, we present a novel tool for electrophysiology: the capacitance clamp provides reliable control over the capacitance of a neuron and thereby opens a new way to study the temporal dynamics of excitable cells.

Thalamocortical circuits from neuroanatomy to mental representations

In highly volatile environments, performing actions that address current needs and desires is an ongoing challenge for living organisms. For example, the predictive value of environmental signals needs to be updated when predicted and actual outcomes differ. Furthermore, organisms also need to gain control over the environment through actions that are expected to produce specific outcomes. The data to be presented will show that these processes are highly reliant on thalamocortical circuits wherein thalamic nuclei make a critical contribution to adaptive decision-making, challenging the view that the thalamus only acts as a relay station for the cortical stage. Over the past few years, our work has highlighted the specific contribution of multiple thalamic nuclei in the ability to update the predictive link between events or the causal link between actions and their outcomes via the combination of targeted thalamic interventions (lesion, chemogenetics, disconnections) with behavioral procedures rooted in experimental psychology. We argue that several features of thalamocortical architecture are consistent with a prominent role for thalamic nuclei in shaping mental representations.

Though the term has many definitions, computational neuroscience is mainly about applying mathematics to the study of the brain. The brain—a jumble of all different kinds of neurons interconnected in countless ways that somehow produce consciousness—has been described as “the most complex object in the known universe”. Physicists for centuries have turned to mathematics to properly explain some of the most seemingly simple processes in the universe—how objects fall, how water flows, how the planets move. Equations have proved crucial in these endeavors because they capture relationships and make precise predictions possible. How could we expect to understand the most complex object in the universe without turning to mathematics? — The answer is we can’t, and that is why I wrote this book. While I’ve been studying and working in the field for over a decade, most people I encounter have no idea what “computational neuroscience” is or that it even exists. Yet a desire to understand how the brain works is a common and very human interest. I wrote this book to let people in on the ways in which the brain will ultimately be understood: through mathematical and computational theories. — At the same time, I know that both mathematics and brain science are on their own intimidating topics to the average reader and may seem downright prohibitory when put together. That is why I’ve avoided (many) equations in the book and focused instead on the driving reasons why scientists have turned to mathematical modeling, what these models have taught us about the brain, and how some surprising interactions between biologists, physicists, mathematicians, and engineers over centuries have laid the groundwork for the future of neuroscience. — Each chapter of Models of the Mind covers a separate topic in neuroscience, starting from individual neurons themselves and building up to the different populations of neurons and brain regions that support memory, vision, movement and more. These chapters document the history of how mathematics has woven its way into biology and the exciting advances this collaboration has in store.

Is there universality in biology?

It is sometimes said that there are two reasons why physics is so successful as a science. One is that it deals with very simple problems. The other is that it attempts to account only for universal aspects of systems at a desired level of description, with lower level phenomena subsumed into a small number of adjustable parameters. It is a widespread belief that this approach seems unlikely to be useful in biology, which is intimidatingly complex, where “everything has an exception”, and where there are a huge number of undetermined parameters. I will try to argue, nonetheless, that there are important, experimentally-testable aspects of biology that exhibit universality, and should be amenable to being tackled from a physics perspective. My suggestion is that this can lead to useful new insights into the existence and universal characteristics of living systems. I will try to justify this point of view by contrasting the goals and practices of the field of condensed matter physics with materials science, and then by extension, the goals and practices of the newly emerging field of “Physics of Living Systems” with biology. Specific biological examples that I will discuss include the following: Universal patterns of gene expression in cell biology Universal scaling laws in ecosystems, including the species-area law, Kleiber’s law, Paradox of the Plankton Universality of the genetic code Universality of thermodynamic utilization in microbial communities Universal scaling laws in the tree of life The question of what can be learned from studying universal phenomena in biology will also be discussed. Universal phenomena, by their very nature, shed little light on detailed microscopic levels of description. Yet there is no point in seeking idiosyncratic mechanistic explanations for phenomena whose explanation is found in rather general principles, such as the central limit theorem, that every microscopic mechanism is constrained to obey. Thus, physical perspectives may be better suited to answering certain questions such as universality than traditional biological perspectives. Concomitantly, it must be recognized that the identification and understanding of universal phenomena may not be a good answer to questions that have traditionally occupied biological scientists. Lastly, I plan to talk about what is perhaps the central question of universality in biology: why does the phenomenon of life occur at all? Is it an inevitable consequence of the laws of physics or some special geochemical accident? What methodology could even begin to answer this question? I will try to explain why traditional approaches to biology do not aim to answer this question, by comparing with our understanding of superconductivity as a physical phenomenon, and with the theory of universal computation. Motor Cortical Control of Vocal Interactions in a Neotropical Singing Mouse

Using sounds for social interactions is common across many taxa. Humans engaged in conversation, for example, take rapid turns to go back and forth. This ability to act upon sensory information to generate a desired motor output is a fundamental feature of animal behavior. How the brain enables such flexible sensorimotor transformations, for example during vocal interactions, is a central question in neuroscience. Seeking a rodent model to fill this niche, we are investigating neural mechanisms of vocal interaction in Alston’s singing mouse (Scotinomys teguina) – a neotropical rodent native to the cloud forests of Central America. We discovered sub-second temporal coordination of advertisement songs (counter-singing) between males of this species – a behavior that requires the rapid modification of motor outputs in response to auditory cues. We leveraged this natural behavior to probe the neural mechanisms that generate and allow fast and flexible vocal communication. Using causal manipulations, we recently showed that an orofacial motor cortical area (OMC) in this rodent is required for vocal interactions (Okobi*, Banerjee* et. al, 2019). Subsequently, in electrophysiological recordings, I find neurons in OMC that track initiation, termination and relative timing of songs. Interestingly, persistent neural dynamics during song progression stretches or compresses on every trial to match the total song duration (Banerjee et al, in preparation). These results demonstrate robust cortical control of vocal timing in a rodent and upends the current dogma that motor cortical control of vocal output is evolutionarily restricted to the primate lineage.

The Desire to Know: Non-Instrumental Information Seeking in Mice

Animals are motivated to acquire knowledge. A particularly striking example is information seeking behavior: animals often seek out sensory cues that will inform them about the properties of uncertain future rewards, even when there is no way for them to use this information to influence the reward outcome, and even when this information comes at a considerable cost. Evidence from monkey electrophysiology and human fMRI studies suggests that orbitofrontal cortex and midbrain dopamine neurons represent the subjective value of knowledge during information seeking behavior. However, it remains unclear how the brain assigns value to information and how it integrates this with other incentives to drive behavior. We have therefore developed a task to test if information preferences are present in mice and study how informational value is imparted on stimuli. Mice are trained to enter a center port and receive an initial odor that instructs them to either go to an informative side port, go to an uninformative side port, or choose freely between them. The chosen side port then yields a second odor cue followed by a delayed probabilistic water reward. The informative port’s odor cue indicates whether the upcoming reward will be big or small. The uninformative port’s odor cue is uncorrelated with the trial outcome. Crucially, the two ports only differ in their odor cues, not in their water value since both offer identical probabilities of big and small rewards. We find that mice prefer the informative port. This preference is evident as a higher percentage choice of the informative port when given a free choice (67% +/- 1.7%, n = 14, p < 0.03), as well as by faster reaction times when instructed to go to the informative port (544ms +/- 21ms vs 795ms +/- 21ms, n = 14, p < 0.001). The preference for information is robust to within-animal reversals of informative and uninformative port locations, and, moreover, mice are willing to pay for information by choosing the informative port even if its reward amount is reduced to be substantially lower than the uninformative port. These behavioral observations suggest that odor stimuli are imparted with informational value as mice learn the information seeking task. We are currently imaging neural activity in orbitofrontal cortex with microendoscopes to identify changes in neural activity that may reflect value associated with the acquisition of knowledge.
