PiVR: An affordable and versatile closed-loop platform to study unrestrained sensorimotor behavior
PiVR is a system that allows experimenters to immerse small animals into virtual realities. The system tracks the position of the animal and presents light stimulation according to predefined rules, thus creating a virtual landscape in which the animal can behave. By using optogenetics, we have used PiVR to present fruit fly larvae with virtual olfactory realities, adult fruit flies with a virtual gustatory reality and zebrafish larvae with a virtual light gradient. PiVR operates at high temporal resolution (70Hz) with low latencies (<30 milliseconds) while being affordable (<US$500) and easy to build (<6 hours). Through extensive documentation (www.PiVR.org), this tool was designed to be accessible to a wide public, from high school students to professional researchers studying systems neuroscience in academia.
Exploring fine detail: The interplay of attention, oculomotor behavior and visual perception in the fovea
Outside the foveola, visual acuity and other visual functions gradually deteriorate with increasing eccentricity. Humans compensate for these limitations by relying on a tight link between perception and action; rapid gaze shifts (saccades) occur 2-3 times every second, separating brief “fixation” intervals in which visual information is acquired and processed. During fixation, however, the eye is not immobile. Small eye movements incessantly shift the image on the retina even when the attended stimulus is already foveated, suggesting a much deeper coupling between visual functions and oculomotor activity. Thanks to a combination of techniques allowing for high-resolution recordings of eye position, retinal stabilization, and accurate gaze localization, we examined how attention and eye movements are controlled at this scale. We have shown that during fixation, visual exploration of fine spatial detail unfolds following visuomotor strategies similar to those occurring at a larger scale. This behavior compensates for non-homogenous visual capabilities within the foveola and is finely controlled by attention, which facilitates processing at selected foveal locations. Ultimately, the limits of high acuity vision are greatly influenced by the spatiotemporal modulations introduced by fixational eye movements. These findings reveal that, contrary to common intuition, placing a stimulus within the foveola is necessary but not sufficient for high visual acuity; fine spatial vision is the outcome of an orchestrated synergy of motor, cognitive, and attentional factors.
The interaction of sensory and motor information to shape neuronal representations in mouse cortical networks
The neurons in our brain never function in isolation; they are organized into complex circuits which perform highly specialized information processing tasks and transfer information through large neuronal networks. The aim of Janelle Pakan's research group is to better understand how neural circuits function during the transformation of information from sensory perception to behavioural output. Importantly, they also aim to further understand the cell-type specific processes that interrupt the flow of information through neural circuits in neurodegenerative disorders with dementia. The Pakan group utilizes innovative neuroanatomical tracing techniques, advanced in vivo two-photon imaging, and genetically targeted manipulations of neuronal activity to investigate the cell-type specific microcircuitry of the cerebral cortex, the macrocircuitry of cortical output to subcortical structures, and the functional circuitry underlying processes of sensory perception and motor behaviour.
Contextual inference underlies the learning of sensorimotor repertoires
Humans spend a lifetime learning, storing and refining a repertoire of motor memories. However, it is unknown what principle underlies the way our continuous stream of sensori-motor experience is segmented into separate memories and how we adapt and use this growing repertoire. Here we develop a principled theory of motor learning based on the key insight that memory creation, updating, and expression are all controlled by a single computation – contextual inference. Unlike dominant theories of single-context learning, our repertoire-learning model accounts for key features of motor learning that had no unified explanation and predicts novel phenomena, which we confirm experimentally. These results suggest that contextual inference is the key principle underlying how a diverse set of experiences is reflected in motor behavior.
Neural mechanisms of proprioception and motor control in Drosophila
Animals rely on an internal sense of body position and movement to effectively control motor behaviour. This sense of proprioception is mediated by diverse populations of internal mechanosensory neurons distributed throughout the body. My lab is trying to understand how proprioceptive stimuli are detected by sensory neurons, integrated and transformed in central circuits, and used to guide motor output. We approach these questions using genetic tools, in vivo two-photon imaging, and patch-clamp electrophysiology in Drosophila. We recently found that the axons of fly leg proprioceptors are organized into distinct functional projections that contain topographic representations of specific kinematic features: one group of axons encodes tibia position, another encodes movement direction, and a third encodes bidirectional movement and vibration frequency. Whole-cell recordings from downstream neurons reveal that position, movement, and directional information remain segregated in central circuits. These feedback signals then converge upon motor neurons that control leg muscles during walking. Overall, our findings reveal how a low-dimensional stimulus – the angle of a single leg joint – is encoded by a diverse population of mechanosensory neurons. Specific proprioceptive parameters are initially processed by parallel pathways, but are ultimately integrated to influence motor output. This architecture may help to maximize information transmission, processing speed, and robustness, which are critical for feedback control of the limbs during adaptive locomotion.
Optogenetic manipulations of cortico-pallidal pathway impact specific aspects of motor behaviour in mice
