Effects of Apolipoprotein A4 on Lipid Metabolism via Sympathetic Regulation
Obesity increases the risks and progression of hypertriglyceridemia, metabolic dysfunction- associated steatotic liver disease (MASLD), and cardiovascular diseases. Previous studies demonstrate that a single injection of apolipoprotein A4 (APOA4) elevates sympathetic neural activity and fatty acid β-oxidation in adipose tissues; and consistent infusion of APOA4 in obese mice fed a high-fat diet lowers fat mass, reduces hypertriglyceridemia, elevates brown adipose tissue thermogenesis, and attenuates steatosis and enhances sympathetic neural activity in the liver. This project hypothesizes that APOA4 reduces hypertriglyceridemia by regulating lipid metabolism through sympathetic stimulation in adipose tissues (Specific Aim 1) and sympathetic action in the liver (Specific Aim 2). The role of sympathetic action via the neurotransmitter norepinephrine and adrenergic receptor-mediated pathways will be investigated, and their necessity in APOA4-mediated lipid metabolism will be tested. A strength of this project is the interdisciplinary collaboration between investigators with established successful collaboration and publications. The project will provide physiological, molecular, and neurochemical mechanisms underlying how APOA4 differentially regulates metabolism through sympathetic activation in various types of adipose tissues and the liver in male and female obese mice. Findings would provide impetus to develop unique, novel, targeted therapeutic applications against hypertriglyceridemia and MASLD. Importantly, this project will expose undergraduates and graduate students to meritorious research, provide students with hands-on biomedical research experience, and strengthen research environment at R15 eligible institutions.
Facilitating the Advancement of Research and Education for Undergraduate Students by Incorporating Laser Scanning Confocal Microscopy (FAREUS-LSCM)
PROJECT SUMMARY/ABSTRACT The University of Puerto Rico at Aguadilla (UPR-Aguadilla) requests funding to acquire a Nikon AX Galvo Confocal Laser Scanning Microscope (LSCM) with a TI2-E inverted platform and a four- laser configuration (405/488/561/640 nm) to establish transformative imaging capabilities at our resource-limited institution serving 96% Pell Grant recipients. This state-of-the-art instrument addresses a critical infrastructure gap, enabling high-resolution fluorescence imaging, live-cell microscopy, and quantitative analysis essential for competitive biomedical research and undergraduate education. The LSCM will directly support four active research projects spanning parasitology (monogenean host-specificity studies), plant pathology (coffee biocontrol development), environmental chemistry (metalloprotein biomarkers), and neuroscience (astrocyte dysfunction in diabetic epilepsy) while integrating into core laboratory courses including Immunology (BIOL 4009) and Undergraduate research courses (BIOL 3108 and QUIM 4999). Our multidisciplinary faculty, in partnership with the Neuroimaging and Electrophysiology Facility (NIEF) Excellence Imaging Center, offers expertise in confocal microscopy, encompassing advanced imaging and specialized sample preparation techniques. This collaboration ensures effective implementation of the technology, sustained technical support, and high-quality training programs that will enhance research productivity and broaden educational impact. The broad, long-term objective is to transform UPR-Aguadilla from a primarily teaching institution into a research-active campus capable of producing graduate-school-ready students equipped with cutting-edge technical skills. Access to advanced confocal microscopy will stimulate new research collaborations, enhance faculty productivity, and provide 30-40 students annually with hands-on experience in modern imaging technologies currently absent from our curriculum. The instrument will strengthen our partnership with the emerging Natural History Museum of Puerto Rico for specimen digitization and support comprehensive outreach programs targeting 25-50 high school students annually through "Seeing Science Up Close" workshops. Expected outcomes include 1- 2 peer-reviewed publications within three years, establishment of 1-2 new institutional collaborations, and measurable enhancement of biomedical research capacity. This investment will significantly advance STEM education and research opportunities at UPR-Aguadilla while expanding access to cutting-edge scientific instrumentation for students pursuing biomedical careers and contributing to the development of skilled researchers in the biomedical sciences.
State-of-the-Art Spike Sorting with SpikeInterface
This webinar will focus on spike sorting analysis with SpikeInterface, an open-source framework for the analysis of extracellular electrophysiology data. After a brief introduction of the project (~30 mins) highlighting the basics of the SpikeInterface software and advanced features (e.g., data compression, quality metrics, drift correction, cloud visualization), we will have an extensive hands-on tutorial (~90 mins) showing how to use SpikeInterface in a real-world scenario. After attending the webinar, you will: (1) have a global overview of the different steps involved in a processing pipeline; (2) know how to write a complete analysis pipeline with SpikeInterface.
Experimental Neuroscience Bootcamp
This course provides a fundamental foundation in the modern techniques of experimental neuroscience. It introduces the essentials of sensors, motor control, microcontrollers, programming, data analysis, and machine learning by guiding students through the “hands on” construction of an increasingly capable robot. In parallel, related concepts in neuroscience are introduced as nature’s solution to the challenges students encounter while designing and building their own intelligent system.
Multi-muscle TMS mapping assessment of the motor cortex reorganization after finger dexterity training
It is widely known that motor learning leads to reorganization changes in the motor cortex. Recently, we have shown that using navigated transcranial magnetic stimulation (TMS) allows us to reliably trace interactions among motor cortical representations (MCRs) of different upper limb muscles. Using this approach, we investigate changes in the MCRs after fine finger movement training. Our preliminary results demonstrated that areas of the APB and ADM and their overlaps tended to increase after finger independence training. Considering the behavioral data, hand dexterity increased for both hands, but the amplitudes of voluntary contraction of the muscles for the APB and ADM did not change significantly. The behavioral results correspond with a previously described suggestion that hand strength and hand dexterity are not directly related as well as an increase in overlaps between MCRs of the trained muscles supports the idea that voluntary muscle relaxation is an active physiological process.
Experience-independent brain development in perception and action systems
From 1D to 5D: Data-driven Discovery of Whole-brain Dynamic Connectivity in fMRI Data
The analysis of functional magnetic resonance imaging (fMRI) data can greatly benefit from flexible analytic approaches. In particular, the advent of data-driven approaches to identify whole-brain time-varying connectivity and activity has revealed a number of interesting relevant variation in the data which, when ignored, can provide misleading information. In this lecture I will provide a comparative introduction of a range of data-driven approaches to estimating time-varying connectivity. I will also present detailed examples where studies of both brain health and disorder have been advanced by approaches designed to capture and estimate time-varying information in resting fMRI data. I will review several exemplar data sets analyzed in different ways to demonstrate the complementarity as well as trade-offs of various modeling approaches to answer questions about brain function. Finally, I will review and provide examples of strategies for validating time-varying connectivity including simulations, multimodal imaging, and comparative prediction within clinical populations, among others. As part of the interactive aspect I will provide a hands-on guide to the dynamic functional network connectivity toolbox within the GIFT software, including an online didactic analytic decision tree to introduce the various concepts and decisions that need to be made when using such tools
A discussion on the necessity for Open Source Hardware in neuroscience research
Research tools are paramount for scientific development, they enable researchers to observe and manipulate natural phenomena, learn their principles, make predictions and develop new technologies, treatments and improve living standards. Due to their costs and the geographical distribution of manufacturing companies access to them is not widely available, hindering the pace of research, the ability of many communities to contribute to science and education and reap its benefits. One possible solution for this issue is to create research tools under the open source ethos, where all documentation about them (including their designs, building and operating instructions) are made freely available. Dubbed Open Science Hardware (OSH), this production method follows the established and successful principles of open source software and brings many advantages over traditional creation methods such as: economic savings (see Pearce 2020 for potential economic savings in developing open source research tools), distributed manufacturing, repairability, and higher customizability. This development method has been greatly facilitated by recent technological developments in fast prototyping tools, Internet infrastructure, documentation platforms and lower costs of electronic off-the-shelf components. Taken together these benefits have the potential to make research more inclusive, equitable, distributed and most importantly, more reliable and reproducible, as - 1) researchers can know their tools inner workings in minute detail - 2) they can calibrate their tools before every experiment and having them running in optimal condition everytime - 3) given their lower price point, a)students can be trained/taught with hands on classes, b) several copies of the same instrument can be built leading to a parallelization of data collection and the creation of more robust datasets. - 4) Labs across the world can share the exact same type of instruments and create collaborative projects with standardized data collection and sharing.
Sensorimotor -independent brain representations in association cortices
How flexible are association cortices? I will present a series of fMRI experiments addressing this question by investigating individuals born without hands, who use their feet as effectors to perform everyday actions. These results suggest that computations in association cortices are abstracted from visuomotor features and experience, similarly to the visual -independence of the association networks in people born blind, highlighting these regions’ ability to compensate for experience in any specific modality. These findings also open new avenues to utilize effector-independence in the action system for motor rehabilitation.
Neuroscience tools for the 99%: On the low-fi development of high-tech lab gear for hands-on neuroscience labs and exploratory research
The public has a fascination with the brain, but little attention is given to neuroscience education prior to graduate studies in brain-related fields. One reason may be the lack of low cost and engaging teaching materials. To address this, we have developed a suite of open-source tools which are appropriate for amateurs and for use in high school, undergraduate, and graduate level educational and research programs. This lecture will provide an overview of our mission to re-engineer research-grade lab equipment using first principles and will highlight basic principles of neuroscience in a "DIY" fashion: neurophysiology, functional electrical stimulation, micro-stimulation effect on animal behavior, neuropharmacology, even neuroprosthesis and optogenetics! Finally, with faculty academic positions becoming a scarce resource, I will discuss an alternative academic career path: entrepreneurship. It is possible to be an academic, do research, publish papers, present at conferences and train students all outside the traditional university setting. I will close by discussing my career path from graduate student to PI/CEO of a startup neuroscience company.
