Brain-wide effects of pontine waves across sleep stages

Mirna Merkler, Shuzo Sakata · FENS Forum 2024

Sleep is an essential process observed throughout the animal kingdom, and it is important for memory consolidation. Coordinated neuronal activity occurs during sleep, characterized by distinct events across different sleep states. Non-rapid eye movement (NREM) sleep features slow oscillations, delta waves, thalamocortical spindles and hippocampal sharp waves-ripples, while REM sleep is dominated by hippocampal theta oscillations and ponto-geniculo-occipital (PGO) waves, or pontine waves (“P-waves”) in rodents. Even though their origin from mesopontine cholinergic and glutamatergic nuclei is kn

Experience-dependent deficits in sharp wave-ripples in 5xFAD mice

Paulina Schnur, Nicole Byron, Tommaso Patriarchi, Shuzo Sakata · FENS Forum 2024

A hallmark of Alzheimer’s disease (AD) pathology is amyloid plaques which can lead to neuroinflammation, cellular dysfunctions as well as altered neuronal activity. Both AD patients and mouse models exhibit abnormal neural oscillations, including altered gamma oscillations, hippocampal sharp wave-ripples (SWRs) and cortical sleep spindles. Although these oscillations play a role in memory formation and consolidation, it remains poorly understood how abnormal oscillations relate to novel experiences. Here we show that deficits in SWRs are experience-dependent in an AD mouse model. We performed
