Acute ischemic stroke, also referred to as a cerebrovascular accident, occurs when localized tissue faces ischemia, leading to an inability to maintain physiological conditions or even resulting in death due to the obstruction or rupture of blood vessels in the brain. The current use of rt-PA for stroke medication has a limited time frame of 3-4.5 hours and is associated with neurotoxicity. Hence, there is an urgent need to explore alternative treatments for ischemic stroke. Cinchonidine, an alkaloid derived from the bark of cinchona, has previously demonstrated anti-inflammatory and antioxida

N,N-Dimethyltryptamine has a protective effect against oxygen-glucose deprivation in a rat primary culture model of the blood-brain barrier

Judit Vigh, Marcell László, Anna Kocsis, Zsófia Hoyk, Fruzsina R. Walter, Attila Szabó, Zoltán Nagy, Mária A. Deli, Sándor Nardai · FENS Forum 2024

Disruption of the blood-brain barrier (BBB) is a key factor in the development of stroke. N,N-dimethyltryptamine (DMT) is a hallucinogen alkaloid widely found in plants and animals. Beneficial effects of DMT was described in a rat stroke model, but BBB functions were not studied. Our aim was to examine the effect of DMT on the functions of the BBB following oxygen-glucose deprivation and reoxygenation (OGDR) in a rat primary BBB co-culture system. To model stroke in vitro, OGD was introduced for 6 hours on the BBB model, followed by reoxygenation for 24 hours with or without the addition of DMT

Organoid technology: A new way to model ischemic stroke

Chiara Giorgi, Giorgia Lombardozzi, Housen Kacem, Fabrizio Ammannito, Michele d'Angelo, Annamaria Cimini · FENS Forum 2024

Ischemic stroke is one of the most serious and widespread neurological emergencies in the world. It is characterized by rapid disruption of brain blood flow, and it can cause irreversible damage to brain cells leading to permanent disability or death. Currently, treatment options are limited and fail to completely prevent or reverse brain damage. It is therefore essential to pursue innovative approaches to improve the prognosis and quality of life of ischemic stroke patients.Brain organoids are three-dimensional models that can reproduce the cellular composition, structure, and neuronal connec

Ischemic stroke results in neuroinflammation. While several studies addressed its molecular mechanism, we know less about the contribution of physical cues to stroke pathomechanism. In the present work, we aimed to address the significance of nervous tissue cytoskeleton rearrangements and changes in its mechanical properties in neuroinflammation. We applied oxygen-glucose deprivation (OGD) on rat organotypic hippocampal cultures (OHCs) as our model of brain ischemia. We observed that OGD significantly increases LHD and NO release from OHCs. HIF-1α protein expression was also significantly incr

Ex vivo direct current stimulation modulates synaptic plasticity under pathological condition with the implication of FKBP51-related signaling

Ching-Hsiang Chang, Ting-Yu Chung, Wan-Jou Lee, Jia-Yi Li, Yu-Feng Chen, Hui-Ching Lin · FENS Forum 2024

Dysregulation of synaptic plasticity was associated with numerous neurological disorders, including ischemic long-term potentiation (iLTP). Direct current stimulation (DCS) changes neuronal plasticity by the weak direct current which induces depolarization and hyperpolarization of resting membrane potential with opposite direction of polarity. FK506-binding protein 51 (FKBP51) is considered an important molecular pathway that modulates synaptic plasticity in such disease models. The correlation of the regulatory effects in plasticity under pathological condition facilitated by DCS and FKBP51 r
