Abstract

Mature spermatozoa are transcriptionally quiescent yet carry a complex payload of long noncoding RNAs (lncRNAs) and small noncoding RNAs (sncRNAs) that can modulate early embryonic transcription. Previous studies showed that a range of environmental perturbations can alter the sperm payload and thereby influence offspring development and propensity for certain neuropsychiatric disease related phenotypes. It is unclear whether a short trauma exposure would alter sperm RNAs and whether RNA binding proteins contribute to the stabilization of RNA payload. We investigated the impact of a one-time acute traumatic foot shock experience on sperm small and long RNA and their dependence on specific proteins present in sperm. We further intersect these data with early embryo transcriptomes obtained by fertilizing naive oocytes with sperm from stressed males to elucidate potential routes of intergenerational information transmission.
