Abstract

Fluorescence microscopy is a cornerstone of modern neurobiology, but its full potential remains limited by the availability of high-performance probes. To address this, we have developed a new generation of protein-based fluorescent probes optimized for super-resolution imaging of synaptic proteins.

Our approach combines rational design and directed protein evolution to create compact, high-affinity binders targeting both generic epitope tags and endogenous proteins. Recently, we successfully engineered novel binders that overcome the size and specificity limitations of existing tools. These probes have been validated and optimized for direct use in advanced imaging applications.

Here, we will showcase our latest results, including the expansion of our toolkit and its successful application in super-resolution microscopy. Our work provides neurobiologists with innovative tools to study protein dynamics in both genetically modified and endogenous contexts, pushing the boundaries of what is achievable with fluorescence microscopy.
