Methods: We engineered a lyophilized agarose–carbomer and polyethylene glycol hydrogel. In vitro experiments assessed the platform's ability to support SVF populations over 10 days. Cell viability was measured using CFDA-SE and propidium iodide staining, while live immunocytochemistry and colocalization microscopy identified specific subpopulations, including ADSCs, endothelial cells, and pericytes.
Results: The SVF-laden hydrogels maintained high cell viability throughout the 10-day culture period. Results confirmed the sustained presence and survival of key regenerative populations, particularly ADSCs, within the hydrogel matrix, suggesting a highly favourable microenvironment for cell retention.
Conclusion: Encapsulating SVF in this specialized hydrogel promotes prolonged in vitro viability. This delivery strategy represents a promising advancement for enhancing the efficacy of regenerative therapies in SCI.
