Early life stress and living in a complex environment: Effects on social hierarchy and stress coping in mice

Abstract

Living in an urban environment demands flexibility in dealing with the complexity of the city, both in public space and in social aspects. Not everyone can keep up in this complex environment and mental health issues are more common in urban compared to rural areas. Adverse early life events might be a risk factor for the transition from mental health to mental illness in an urban environment. This study aimed to investigate how stressful experiences in sensitive developmental periods early in life influence later life coping in a demanding social environment. We subjected mice to limited nesting and bedding to induce early life stress (ELS). In adulthood, mice were housed in a complex environment for 10 days during which social hierarchy was measured. Two weeks later, mice were subjected to predator odour, followed by a choice task where they could choose between access to their cagemate or access to a shelter. We found that ELS did not significantly influence social hierarchy or choice behavior after stress. Male mice who have experienced complex housing show an increase in freezing time and a decrease in rearing frequency when exposed to predator odour. During the choice task, we found that a past complex housing experience inhibited contact initiation in male mice. Our findings suggest that an adult experience with complex housing influences subsequent stress coping in males, while early life stress does not strongly impact hierarchy or stress coping in both sexes.
