Prediction and prevention of compulsive behaviors by closed-loop optogenetic recruitment of striatal interneurons

Abstract

Dysregulation in the cortico-striatal circuits has been shown to be related to pathologic repetitive behaviors but the underlying dysfunctional mechanisms at the microcircuit level remain under investigation. A key element that regulate the striatal output iare striatal parvalbumin (PV) interneurons which exert a powerful feedforward inhibition on medium spiny neurons. To investigate the role of PV interneurons in regulating aberrant striatal activity observed in repetitive behaviors, we used the Sapap3 mutant mice, which exhibit excessive self-grooming. Using optogenetics activation of PV interneurons in the centromedial striatum, we were able to reduce compulsive behavior to normality. Moreover, we found that an increase in power of 1-4 Hz oscillations in the orbitofrontal cortex temporally predicted grooming initialization. We then used a combination of extracellular signal acquisition, optogenetics manipulation, and online data processing to predict grooming onset and deliver optogenetic stimulation of striatal PV interneurons in a closed-loop design. We found that both, closed-loop optogenetics and continuous open loop stimulation robustly decreased excessive grooming. Our closed-loop stimulation design allowed us to demonstrate that recruitment of PV interneurons was particularly important at the onset of the compulsive events to regulate their expression. Moreover, we have provided an experimental framework that allows the prediction of grooming behavior, which pave the way novel therapeutical approach in the field of deep brain stimulation.
