Abstract

Although tobacco-free nicotine pouches are often framed as lower-risk alternatives, their toxicological profiles are still being characterized and long term health effects remain uncertain. These pouches and are available in a wide range of flavours and contain nicotine, which possesses strong addictive properties, and also complex flavour compositions which have shown toxic effects on previous in vitro studies. But studies on neuronal cells are missing. In this study, human SH-SY5Y cells are treated with extracts from a tobacco-free nicotine pouch, a tobacco- and nicotine-free pouch and a tobacco pouch to evaluate the products cytotoxic properties and potential effects on cell viability in neuronal cells, using LDH and calcein-AM assays. The results suggested that exposure to all three extracts was accompanied by detectable differences in both LDH and calcein intensity. Under the conditions used, changes could be observed across the three pouch extracts, indicating that the observed responses varied between products. In conclusion, across the assays applied, the three products were associated with differences in the measured readouts in SH-SY5Y cells. As the mechanisms underlying these observations remain undetermined, these findings may be relevant to investigate further in future studies.
