Abstract

Epidemiological studies indicate that women are more likely to develop Alzheimer’s disease (AD) than men. Consistent with these sex-specific effects, several AD mouse models report greater amyloid pathology in females. To our knowledge, only one study has reported early cognitive impairment associated with higher amyloid burden in females compared to males in the Tg2576 (APPswe) model. The mechanisms underlying these sex differences remain poorly characterized. In this project, we aim to investigate the central and peripheral mechanisms that may contribute to sex differences in the progression of AD. Memory performance was assessed in male and female Tg2576 mice and non-transgenic counterparts at 4 and 17 months of age using a topological pattern separation task. This task assesses the animal’s ability to detect subtle topological changes and critically depends on intact adult hippocampal neurogenesis. After behavioral testing, mice were sacrificed and brain and intestinal tissue were collected. Adult hippocampal neurogenesis and inflammation in the central nervous system and periphery will be assessed. By combining behavioral, neurogenic, and inflammatory analyses in the brain and gut, we aim to establish a correlation between cognitive performance, hippocampal integrity and inflammatory profiles. This study will also determine whether these relationships differ between sexes in the Tg2576 mouse model of AD.
