Histone deacetylase (HDAC) family proteins play important roles in pathological alterations of Alzheimer’s disease (AD). There has been keen interest in developing small molecule inhibitors of HDACs as potential therapeutics of AD. Presently, little is known about the role of HDAC11 in AD and whether HDAC11 can be a potential drug target for AD. Recently, we discovered PB94 as our lead small molecule inhibitor of HDAC11 exhibiting strong potency and selectivity against HDAC11 over other HDAC isoforms, which warranted further optimization and investigation for AD. Here, we optimized PB94 and an

Examining the effects of galectin-3 on neurons and glia

Jun Ong, Sofia Savastano, Francis Szele · FENS Forum 2024

Galectin-3 is upregulated in Alzheimer's Disease (AD) and has been shown to promote amyloid beta (Aβ) oligomerisation and plaque formation. However, little is known about the effect of galectin-3 on neural cells. Our work aims to determine the effect of galecitn-3 on neurogenesis and on glial clearance of Aβ. Neurospheres generated from primary murine neural stem cells were treated with galectin-3 or the galectin-3 inhibitor TD139. Proliferation, as determined by neurosphere size after 5 days, was decreased in neurospheres treated with galectin-3. Differentiation, as quantified by proportion o
