Immediate-early genes: Key influencers in genetic networks and their role in Alzheimer’s disease

Margarita Zachariou, Eleni M. Loizidou, George M. Spyrou · FENS Forum 2024

Immediate-early genes (IEGs) are a class of activity-regulated genes (ARGs) that are transiently and rapidly activated in the absence of de novo protein synthesis in response to neuronal activity. We explored the role of IEGs in genetic networks to pinpoint potential drug targets for Alzheimer’s disease (AD). Using a combination of network analysis and genome-wide association study (GWAS) summary statistics we show that (1) IEGs exert greater topological influence across different human and mouse gene networks compared to other ARGs, (2) ARGs are sparsely involved in diseases and significantly

Whole brain mapping of engram distribution and stability

Miguel Fuentes Ramos, Marta Alaiz-Noya, Federico Miozzo, Alba Vieites-Prado, Nicolas Renier, Angel Barco · FENS Forum 2024

Both classical activity mapping based on the induction of activity-regulated genes and novel genetic methods to label and control the activity of neurons indicate that memories are deposited in discrete neuronal ensembles in the hippocampus, amygdala and other brain regions involved in memory formation. However, the creation of a brain-wide map for engram formation and stability has proven to be a challenging endeavor that has been only partially solved. Among the methods to label active neurons, the Targeted Recombination in Active Populations v2 (TRAP2) mouse strain sticks out for enabling t
