Astrocyte-based interleukin-2 gene therapy in temporal lobe epilepsy

Evelien Hendrix, Ilse Smolders, Matthew Holt · FENS Forum 2024

Epilepsy, a neurological disorder affecting approximately 60 million individuals globally, poses a significant medical challenge with a substantial unmet need. Temporal lobe epilepsy is the most prevalent form, often triggered by traumatic incidents or status epilepticus (SE), resulting in hippocampal damage characterized by neuronal loss, astro- and microgliosis and neuroinflammation preceding the spontaneous recurrent seizures. Recently, an AAV-PHP.B-GFAP-interleukin-2 (IL-2) gene therapy exhibited impressive neuroprotective and anti-inflammatory effects in mouse models of traumatic brain in

Exacerbation of tauopathy following mild and repeated spinal contusions in the hTauP301S mouse model

Nicolas Halloin, Kathleen De Swert, Lindsay Sprimont, Valéry Bielarz, Kunie Ando, Karelle Leroy, Charles Nicaise · FENS Forum 2024

Chronic traumatic encephalopathy is a tauopathy resulting from repeated moderate-to-severe traumatic injuries onto the brain, leading to Tau protein abnormal posttranslational modifications, mislocalization, and deposits. Although the spinal cord shares a lot of structural and cellular similarities, most studies of CNS traumatic injuries were conducted onto the brain. In the present project, we emitted the hypothesis that repeated mild contusions onto the spinal cord could similarly exacerbate the tauopathy in a transgenic mouse expressing an aggregation-prone human mutated tau protein (hTauP3
