Asymmetric metabolism controls the developing axon complexity in post-mitotic neurons

Fumi Suomi, Anna Rappe, Frédéric Clotman, Thomas McWilliams · FENS Forum 2024

Aims: Metabolic profile impacts on tissue development including neurons in brain, yet the full repertoire of metabolic signals that shape developing neuronal architecture remain mysterious. Our aim is to clarify the metabolic crosstalk that coordinates the early acquisition of the neurite complexity in developing neurons. Methods: To investigate the factors involved in metabolic switch, we used transcriptomics analysis on the sensory neurons cultured in different metabolic demand. IPA analysis was used to interpret the transcriptomics data. We applied MEME-ChIP method on comprehensive motif an

Dynamic TRPV2 localization by focal mechanical stimulation enhances growth cone motility

Koji Shibasaki, Shouta Sugio · FENS Forum 2024

We previously reported that TRPV2 can be activated by mechanical stimulation which enhances axonal outgrowth in developing neurons (Shibasaki et al., J. Neurosci. 2010). However, the molecular mechanisms governing the contribution of TRPV2 activation to axonal outgrowth remain unclear. In this study, we examined this mechanism. Overexpression of TRPV2 enhanced axonal outgrowth in a mechanical stimulus-dependent manner. Accumulation of TRPV2 at the cell surface was 4-fold greater in the growth cone compared to the soma. In the growth cone, TRPV2 is not static, but dynamically accumulates (withi

Modelling chemotherapy-induced peripheral neuropathy on-a-chip

Xandor Spijkers, Georgia Avramidou, Wouter Strijker, Mary McFarlane, Catherine Rodger, Jay Harper, Luke Masterson, Nienke Wevers · FENS Forum 2024

Peripheral neuropathy is characterized by damage to the peripheral nerves, leading to symptoms such as numbness, tingling, and pain in the extremities. While peripheral neuropathy can be caused by several factors, a common cause is toxicity from chemotherapeutic drugs. We studied chemotherapy-induced peripheral neuropathy (CIPN) using the OrganoPlate, a microfluidic cell culture platform that allows culture of 40 chips in parallel. Human iPSC-derived peripheral neurons were embedded in extracellular matrix gel and seeded into the OrganoPlate, after which they extended their axons into an adjace
