The chromatin remodeler CHD7 acts as a chromatin hub coordinating differentiation of multiple cell lineages during hippocampal development

Chromatin remodelling plays an essential role in development, for instance by controlling gene expression during cell differentiation. Mutations of chromatin remodellers are often implicated in developmental diseases such as CHARGE syndrome, in which a heterozygous mutation of the chromatin remodeller CHD7 is the major driver of the disease. To investigate the role of Chd7 during brain development, we used a dorsal telencephalon-specific Chd7 loss-of-function mouse model and discovered Chd7 acts as a multi-lineage coordinator. Histological analysis of Chd7 mutant mice shows that upon ablation

Postnatal Cajal-Retzius cell ablation induced learning deficits in young adult mice

Ingvild Glærum, Rob Machold, Giulia Quattrocolo · FENS Forum 2024

The hippocampus is a brain structure renowned for its role in memory formation, learning and the representation of space. Hippocampal circuitry is comprised of a diverse set of excitatory and inhibitory neurons, including a transient population of reelin+ glutamatergic Cajal-Retzius neurons (CR). In contrast to neocortical CR-cells, which disappear soon after birth, hippocampal CR-cells persist for an extended period during postnatal development. These CR-cells are actively integrated into the hippocampal microcircuit; however, their influence on postnatal hippocampal development and function

The role of GABAergic inputs and chloride transporters in the activation and death of different Cajal-Retzius neuron subpopulations

Federico De Rosa, Ahd Abusaada, Heiko J. Luhmann, Werner Kilb, Anne Sinning · FENS Forum 2024

Cajal-Retzius neurons (CRNs) are among the first-born neurons in the neocortex and are critically involved in cortical layering during their transient lifetime. The molecular mechanisms behind early death of CRNs are still unclear but apoptosis is regulated by neuronal activity. During early development, CRNs are embedded into immature circuits where they mostly receive GABAergic inputs. Through immunohistochemistry and high-resolution confocal imaging, we quantified GABAergic synapse localizations across cortical layers during development in early postnatal mice and compared the GABAergic pro
