The coupling of voltage-gated calcium channels (VGCCs) and vesicular calcium sensors for neurotransmitter release is crucial for fast and efficient synaptic transmission. At parallel fiber (PF)-Purkinje cell (PC) synapses in mouse cerebellum, the coupling tightens during synaptic maturation in parallel with switching of splicing isoforms of the P/Q-type VGCC (Cav2.1) EF-hand from EFb to EFa. The Cav2.1[EFa] isoform has been reported to have a tighter Ca2+ influx-sensor coupling distance than the Cav2.1[EFb] isoform in hippocampal pyramidal neurons. Thus, the shift in the splicing Cav2.1-EF han

Cav2.1 channels are critically important for fast synaptic transmission at central synapses. The two Cav2.1 isoforms Cav2.1[EFa] and Cav2.1[EFb] are generated by alternative splicing of two mutually exclusive exons, 37a and 37b. Both isoforms are expressed throughout the adult brain, whereas Cav2.1[EFb] predominates at early stages of development. Cav2.1[EFa] supports synchronous release and short-term synaptic depression, whereas Cav2.1[EFb] boosts asynchronous release and short-term facilitation. We investigated whether the two isoforms were distinctly located at excitatory nerve terminals r
