A hallmark of nervous system aging is a decline of white matter volume and function, but the underlying mechanisms leading to white matter pathology are unknown. In the present study, we found age-related alterations of oligodendrocyte cell state with a reduction in total oligodendrocyte density in aging murine white matter. Using single-cell RNA-sequencing, we identified interferon (IFN)-responsive oligodendrocytes, which localize in proximity to CD8+ T cells in aging white matter. Absence of functional lymphocytes decreased the number of IFN-responsive oligodendrocytes and rescued oligodendr

Neuroimmune dysregulations strongly contribute to the pathophysiology of Alzheimer's Disease (AD). Because pathogens shape the immune system over lifetime, their association with AD has been suggested. Increasing evidence supports a role for the prevalent, brain-persisting parasite Toxoplasma gondii (Tg) in chronic neurological diseases. Yet, the contribution of Tg chronic infection on AD clinical progression and pathogenesis remains unexplored. Considering the temporal relationship between natural Tg infection and AD occurrence, it seems legitimate to evaluate the effects of a seemingly silen

Alzheimer´s disease (AD) progression has been recently associated to the infiltration of CD8+ T cells into disease-affected brain parenchyma, where they tightly associate with microglial and neuronal structures. However, the functional role of CD8+ T cells in the AD brain is still elusive. Therefore, we investigated the impact of short-term CD8+ T cell ablation on neuroinflammation in transgenic AD mice (APP/PS1).CD8+ T cells ablation was performed via intraperitoneal injections of anti-CD8 antibody for three days in two-year-old APP/PS1 and wild-type male mice. Control groups received the res
