Impact of chronic stress and cortisol on hippocampal neuroplasticity: Implications for depression

Joseph Serrano, Kathleen Hegadoren, Nikolai Malykhin · FENS Forum 2024

Chronic psychosocial stress is a major etiological risk factor for major depressive disorder. Preclinical studies have shown that chronic stress can suppress adult neurogenesis in the dentate gyrus (DG). The primary goal of the present study was to test whether chronic stress and cortisol level translate into detectable changes in DG in humans. We recruited 55 participants without a history of psychiatric disorders. Clinical symptoms, perceived stress, and chronic stressors were rated on the Mood & Anxiety Disorders Symptom Questionnaire, Perceived Stress Scale, and Trier Inventory for Chr

A novel role for LSD1 splicing modulation in homeostatic adaptation to chronic stress

Arteda Paplekaj, Chiara Forastieri, Elena Romito, Andrea de Donato, Sara Testa, Emanuela Toffolo, Elena Battaglioli, Francesco Rusconi · FENS Forum 2024

The stress response serves as a protective mechanism to address daily challenges, yet it's crucial for it to cease when the threat subsides, as prolonged stress engagement links to higher vulnerability to neuropsychiatric disorders.Lysine Specific Demethylase 1 (LSD1) is an epigenetic enzyme participating in the CoREST/HDAC2 corepressor complex regulating neuronal plasticity-related gene transcription. In the mammalian brain, LSD1 activity is modulated by its neurospecific splicing isoform neuroLSD1, lacking co-repressive activity as long as the ability to recruit corepressor partners. Numerou

Stress promotes peripheral immune interactions at the brain endothelium

Johana Alvarez, Flurin Cathomas, Kenny L Chan, Lyonna F Parise, Aarthi Ramakrishnan, Molly Estill, Scott J Russo · FENS Forum 2024

Chronic psychosocial stress is a significant risk factor for the development of stress-associated psychiatric disorders, including major depressive disorder, which causes profound debilitation and has increasing worldwide prevalence. Pre-clinical and clinical studies have linked peripheral immune system alterations to stress-related disorders, such as elevated levels of proinflammatory immune cells and cytokines in circulation. Endothelial cells are critical components of the blood-brain barrier (BBB), as they interface directly with immune cells and their released factors, which can enter the
