Implantation and integration of human neural stem cells (hNSCs) to mouse host brains has been reported by previous studies. We are interested in generating a pre-organised construct from hNSCs that contains progenitors with different cerebral cortical layer identity using 3D printing technologies (Jin et al., Nat Commun 14, 5986 (2023). We optimised a sequential isolation protocol of lower and upper cortical layer specific hNSCs generated from human pluripotent stem cells (hPSCs). We confirmed the ability of these cells to generate neurons of the upper or lower cortical layers both in vitro an

Levels of serotonin during brain development important for the functional interplay of instructing transmitter systems

Marta Samina, Thibault Merkelijn, Sabrina Hanswijk, Lidiane Garcia, Josefine Witteveen, Sara Migliarini, Massimo Pasqualetti, Judith Homberg, Sharon Kolk · FENS Forum 2024

The prefrontal cortex (PFC) is responsible for higher cognitive tasks, and its ‘malfunction’ is shared across serotonin (5-HT)-related neuropsychiatric disorders and often find its origins during development. Proper cortical development is directed by a sequence of intrinsic ( transcription factors, local placental-derived 5-HT) and extrinsic (instructing 5-HT and catecholaminergic afferents) events. Recently, our group has shown a functional interplay between the 5-HT and catecholaminergic prefrontal projection systems during development leading to a distortion in PFC cytoarchitecture. Lack o
