The role of GABAergic inputs and chloride transporters in the activation and death of different Cajal-Retzius neuron subpopulations

Federico De Rosa, Ahd Abusaada, Heiko J. Luhmann, Werner Kilb, Anne Sinning · FENS Forum 2024

Cajal-Retzius neurons (CRNs) are among the first-born neurons in the neocortex and are critically involved in cortical layering during their transient lifetime. The molecular mechanisms behind early death of CRNs are still unclear but apoptosis is regulated by neuronal activity. During early development, CRNs are embedded into immature circuits where they mostly receive GABAergic inputs. Through immunohistochemistry and high-resolution confocal imaging, we quantified GABAergic synapse localizations across cortical layers during development in early postnatal mice and compared the GABAergic pro

Spatial proteomics reveals distinct protein patterns in cortical migration disorders caused by LIN28A overexpression and WNT activation

LIN28A is an RNA-binding protein and stem cell marker, and its overexpression was described in rare malignant brain tumours of early childhood. CTNNB1 is an effector of the WNT pathway which plays important roles during development and activating mutations have been reported in malignant brain tumours. In order to investigate the interplay of these oncogenic proteins during embryonal brain development, we overexpressed both in murine neural progenitor cells in vivo. The sole overexpression of either LIN28A, stabilized Ctnnb1 or the combination of both in hGFAP-positive forebrain precursor cells
