Lilii Bulbus (Lilium lancifolium Thunberg) has a proneurogenic effect on the hippocampus. However, its effects on epilepsy and associated pathological features remain unknown. In this study, we investigated the antiseizure effects of a water extract of Lilii Bulbus (WELB) in mouse model of pentylenetetrazol (PTZ)-induced seizure. Mice were injected with PTZ once every 48 h until full kindling was achieved. WELB (100 and 500 mg/kg) was orally administered once daily before PTZ administration and during the kindling process. We found that WELB treatment protected against PTZ-induced low seizure

Engineering capsid-variant AAVs for selective gene delivery to dentate gyrus granule cells in the hippocampus

Severine Deforges, Jihad El Abdari, Claire Domenger, Dirk Grimm, Christophe Mulle · FENS Forum 2024

In order to interrogate the connectivity and function of local hippocampal circuits, there is a strong need to develop viral gene transfer tools which selectively target distinct neuronal populations. Here we have developed a new recombinant-capsid AAV variants (rAAV) which can infect Dentate granule cells (DGC) with high efficiency and very good selectivity. We have used an AAV-peptide library in which each rAAV displays a random 9 amino acids peptide inserted into exposed loops of the AAV9 capsid. The library was injected in vivo by steretaxy in the mouse DG. After 3 rounds of in vivo selecti

Long-term potentiation at the medial perforant path-granule cell synapse requires protein synthesis

Ala Alhalholy, Coralie Berthoux, Pablo Castillo · FENS Forum 2024

One of the most fascinating properties of the brain is its ability to adjust behaviors, which typically results from experience-dependent modifications of neuronal circuits. Synaptic plasticity refers to changes in synaptic strength at pre-existing synapses induced by transient activity. By modifying neural circuit flow, synaptic plasticity is believed to play a central role in the capacity of the brain to incorporate new experiences into persistent memories. Long-term potentiation has emerged as a prominent mechanism associated with learning and memory. Protein synthesis is well-recognized as

The extracellular matrix glycoprotein reelin is best known for orchestrating migration and positioning of cortical neurons during embryonic development. The reeler mutant mouse, lacking reelin expression, displays severe migration defects and aberrant dendritic development of cortical neurons. Less is known about the consequences of postnatally induced reelin deficiency in conditional reelin knockout (reln cko) mice. Postnatally induced reln cko was shown to entail functional deficits of neuronal communication, while morphological deficits of neurons have not been reported so far. Here, we inv

Synaptopodin regulates denervation-induced plasticity at hippocampal mossy fiber synapses

Pia Kruse, Gudrun Brandes, Hanna Hemeling, Zhong Huang, Christoph Wrede, Jan Hegermann, Andreas Vlachos, Maximilian Lenz · FENS Forum 2024

Denervation of brain regions is a common aspect of neurological diseases caused by traumatic brain injury or demyelination. Nevertheless, the mechanisms underlying lesion-induced reorganization of the affected brain regions is a matter of ongoing investigation.Using an entorhinal cortex lesion in mouse organotypic entorhino-hippocampal tissue cultures, we investigated denervation-induced adaptations in dentate granule cells, CA3 pyramidal neurons and their monosynaptic connection – the mossy fiber synapse. For functional and structural analysis of these cells, we employed single and paired who
