Adenosine triggers astrocyte reactivity to drive the pathogenesis of sepsis-associated encephalopathy

Qilin Guo, Davide Gobbo, Na Zhao, Qing Liu, Li-Pao Fang, Tanja M. Gampfer, Markus R. Meyer, Xianshu Bai, Anja Scheller, Frank Kirchhoff, Wenhui Huang · FENS Forum 2024

Molecular signals mediating systemic inflammation entering the brain parenchyma to induce sepsis-associated encephalopathy (SAE) remain elusive. In this study, we demonstrate that peripheral lipopolysaccharide (LPS)-induced systemic inflammation could rapidly increase the plasma adenosine concentration, peaking at 6 hours post LPS injection (hpi). Using in vivo 2P-LSM imaging of adenosine sensor GRABAdo, we observed elevated adenosine levels in the cortex of awake mice, also peaking at 6 hpi. These concomitant pathological events strongly suggest adenosine in the circulation can join forces to

Cell-type specific targeting of JNK inhibitor in brain using systemic AAVs elicits differential effects on affective behaviours in mice

Anniina Jokela, Nikita Tiwari, Jismi John, Prasannakumar Deshpande, Soroush Abyari, Eleanor Coffey · FENS Forum 2024

Upstream regulators on the c-Jun amino-terminal kinase (JNK) pathway are associated with neuropsychiatric disorders such as schizophrenia, bipolar, and autism. However, the mechanism of action of the JNK pathway in the context of these diseases is unknown. We previously identified that inhibition of JNK in dentate gyrus granule cells of mouse hippocampus induces a low anxiety- and low depressive-like phenotype (Mohammad et al., Molecular Psychiatry, 2018). However, whether JNK pathway activity in other brain regions contributes to these behaviours is not known. Here we use cell restrictive exp

High-fat diet mouse model: Effect on anxious/depressive-like phenotype and the impact of gender

Júlia Senserrich Guerrero, Camilla Samuelli, Elena Castro, Alvaro Diaz, Albert Adell, Angel Pazos, Fuencisla Pilar-Cuellar · FENS Forum 2024

Major Depressive Disorder (MDD) is a high-prevalence neuropsychiatric condition characterized by depressed mood and anxiety. The risk of suffering MDD is higher in people with obesity and metabolic dysregulation. We will evaluate the effect of a high-fat diet (HFD) and the impact of gender on behavioural, neurochemical and molecular markers associated with depression/anxiety.Male and female C57BL/6 mice, 6-weeks old, were given a 7-week HFD (60% fat). The behavioural tests performed were: open field test (OFT), novelty-suppressed feeding (NSF) test, sucrose preference test (SPT), and tail susp

The role of prolactin-releasing peptide (PrRP) in the depressive-like behaviours in rats

Anita Kovács, Evelin Szabó, Kristóf László, László Lénárd, Zsuzsanna Tóth, Balázs Gaszner, Viktória Kormos, Dóra Zelena · FENS Forum 2024

Several data suggest that the RFamide peptides - especially the prolactin-releasing peptide (PrRP) – play a role in the regulation of stress responses, the disturbances of which can cause many diseases, including depression. PrRP was identified as an endogenous ligand of the GPR10 receptor, but can also bind to the NPFF2 receptor. Here we would like to reveal the association between depressive-like behaviour and changes in the PrRP regulation. Fifteen minutes forced swimming test (FST) was used to induce depression-like symptoms in male Wistar rats. On the next day during a 6 min test we ident
