Aging of the blood-brain barrier: Metabolic profiling of mouse brain endothelial cell senescence

Hari Baskar Balasubramanian, Ammar Tahir, Dominik Kahr, Florian Haage, Marietta Zille · FENS Forum 2024

The blood-brain barrier (BBB) separates the peripheral circulation from the brain. Brain endothelial cells (BECs) line the inner lumen of blood vessels in the brain and express tight junction proteins, that prevent the leakage of molecules between the cells into the brain, and efflux transporters that actively pump molecules out of the brain. As we age, the BBB becomes leaky and dysfunctional, leading to cerebrovascular diseases and dementia. We here investigated how BECs become senescent, a major feature of aging, and change their metabolic profile. First, we demonstrate that both the chemoth

Understanding the role of microglia in ‘chemofog’ to resolve chemotherapy-induced cognitive impairment

Agnese Cherubini, Giorgia Scaringi, Bernadette Basilico, Davide Ragozzino, Ingrid Reverte · FENS Forum 2024

Cancer survivors treated with chemotherapy experience persistent memory deficits, commonly known as ‘chemofog’. Currently there are no effective treatments for chemotherapy-induced cognitive impairment, and the underlying molecular mechanisms are poorly understood. Recent evidence suggests that chemotherapy induces chronic microglia reactivity, the brain’s resident immune cells, which could drive alterations in neuroplasticity mechanisms ultimately causing memory impairment. In this study we want to investigate the role of microglia in chemotherapy-induced learning and memory impairment in a r
