Capacitance clamp - artificial capacitance in biological neurons via dynamic clamp

Paul Pfeiffer · Schreiber lab, Humboldt University Berlin, Germany

A basic time scale in neural dynamics from single cells to the network level is the membrane time constant - set by a neuron’s input resistance and its capacitance. Interestingly, the membrane capacitance appears to be more dynamic than previously assumed with implications for neural function and pathology. Indeed, altered membrane capacitance has been observed in reaction to physiological changes like neural swelling, but also in ageing and Alzheimer's disease. Importantly, according to theory, even small changes of the capacitance can affect neuronal signal processing, e.g. increase network

The subiculum is one of the most important brain structure in the hippocampal formation which has an essential role in forwarding the hippocampal information to different cortical and subcortical regions. Pyramidal cells are the principal cell type in the subiculum and based on their firing dynamics, they can be divided into regular and bursting phenotypes. Previously, functional characterization of these cells was mainly performed by using conventional electrophysiological methods (current steps). However, our research group uses advanced methods (dynamic clamp) to expose neurons to physiolog

Multifactorial approach is needed to unravel the maturation phases of human neurons derived from induced pluripotent stem cells

Maissa Ben mahmoud, Anikó Rátkai, Krisztina Bauer, Norbert Bencsik, Attila Szücs, Katalin Schlett, Krisztián Tárnok · FENS Forum 2024

Neurons derived from induced pluripotent stem cells (h-iPSC-Ns) provide an invaluable model for studying the physiological aspects of neuronal development and diseases. However, multiple studies have also demonstrated that h-iPSC-Ns exhibit a high degree of functional and epigenetic diversity. Due to the imprecise characterization and significant variation among the currently available maturation protocols, it is essential to establish a set of criteria to standardize models and accurately characterize and define the developmental properties of neurons derived from iPSCs. In this study, we con
