Experience and reactivation status determine engram synapse structural connectivity

Panthea Nemat, Rolinka J. van der Loo, August B. Smit, Sabine Spijker, Priyanka Rao-Ruiz · FENS Forum 2024

Memory engram cells, activated by learning, undergo input-specific augmentation in their synaptic connectivity (structure, strength and efficacy) to facilitate memory storage and recall. However, only a subset of these cells is reactivated upon retrieval, indicating potential differences in engram cell synaptic connectivity. In this study, we investigated structural synaptic connectivity differences between reactivated and non-reactivated hippocampal CA1 engram cells under conditions of neutral and aversive contextual memory valence. A combination of eGRASP (enhanced green fluorescent protein

Neuronal circuit for multisensory integration in higher visual cortex

Mio Inoue, Yuta Tanisumi, Daisuke Kato, Nanami Kawamura, Akari Hashimoto, Ikuko Takeda, Etsuko Tarusawa, Hiroaki Wake · FENS Forum 2024

Multisensory integration is a sensory processing mechanism that integrate input information from different sensory modalities, occurring during sensory reception and perception. However, the underlying neuronal circuit to integrate sensory information is still unknown. We featured anterolateral visual area (AL), which has axonal projection from primary sensory cortexes, and try to clarify the mechanism underpinning sensory information processing and integration in AL. We first visualized neuronal response to sensory input (whisker stimulation and auditory representation) in AL by in vivo Ca2+

The parvalbumin-interneuron mediated modulation of a memory engram in the context of Alzheimer’s disease

Thije Willems, Harm Krugers, Helmut Kessels, Sylvie Lesuis · FENS Forum 2024

Neuronal network imbalance and synaptic dysfunction both critically contribute to cognitive decline in AD. Network imbalance is increasingly recognized as an early characteristic of AD, which might precede and drive clinical symptoms. Early alterations in parvalbumin (PV+) interneurons contribute to this network dysfunction in AD mouse models. However, to what extent this shapes how memories are stored within the brain and drives cognitive decline remains to be determined. Our objective is to investigate synaptic, local, and network imbalances in AD, as well as their mutual dependence. We used

The proteomic architecture of the synaptic engram supporting context memory

Biswajit Moharana, Panthea Nemat, Renee Pullen, Anna Gradl, Remco Klaassen, Cora Chadick, Rolinka van der Loo, Yvonne Gouwenberg, Frank Koopmans, Juan Garcia Vallejo, Michel van den Over, August Smit, Priyanka Rao-Ruiz · FENS Forum 2024

Sparsely distributed hippocampal CA1 engram cells, activated at the time of learning and receiving input from the CA3, are critical to contextual memory consolidation. This manifests as an augmentation of synaptic connectivity between these hippocampal subfields. Enhanced green fluorescent protein (GFP) reconstitution across synaptic partners (eGRASP) facilitates structural and electrophysiological analysis of input-specific synapses between engram cells. However, the protein landscape shaping and maintaining this enhanced engram synaptic connectivity remains unknown. Global proteomic analysis

Stress susceptibility-dependent synaptic plasticity onto anterior cingulate cortex

Sabahaddin Taha Solakoglu, Evren Erdener, Olga Gliko, Alp Can, Uygar Sumbul, Emine Eren-Kocak · FENS Forum 2024

Aims: Investigation of differences in synaptic inputs from ventral tegmental area (VTA), ventral hippocampus (VH) and basolateral amygdala (BLA) on anterior cingulate cortex(ACC) neurons across all cortical layers between stress susceptible (SS) and stress resilient (SR) mice. Methods: Projections from VTA, VH and BLA excitatory neurons were labeled with cyan, green and yellow pre-eGRASP and ACC pyramidal neurons were labeled with post-eGRASP and TagRFP-T. Three weeks later, 10 mice were exposed to chronic social defeat stress for 10 days and on 11th day mice were divided into SS and SR groups
