Modulation of mouse hippocampal lipidomic signature by FTY720, a sphingosine-1-phosphate analogue used for treating relapsing-remitting multiple sclerosis

FTY720 (fingolimod), a synthetic sphingosine 1-phosphate (S1P) analogue, is a drug employed for treating relapsing-remitting multiple sclerosis in diverse age groups. With the ability to cross the blood-brain barrier, FTY720 gradually accumulates within lipid-rich areas of the central nervous system (CNS) by integrating into phospholipid membranes. Consequently, FTY720 can enhance cell membrane fluidity, influencing glial cells and neuronal populations involved in behaviour regulation. This study investigated the impact of chronic FTY720 administration (3.3 μg/ml in ad libitum drinking water f

Sex-dependent BDNF-mediated effects of Fingolimod on the architecture of mouse hippocampal neurons

Aiswaria Lekshmi Kannan, Charlotte Tacke, Martin Korte, Marta Zagrebelsky · FENS Forum 2024

Brain derived neurotrophic factor (BDNF) plays a crucial role in regulating synaptic plasticity and neuronal architecture through complex downstream signalling cascades mediated via receptor Tyrosine Kinase B (TrkB). Impaired BDNF/TrkB signalling has been associated with several neurological disorders. A treatment with Fingolimod has been shown in vitro and in vivo to promote BDNF synthesis and release and exert BDNF-dependent neuroprotection. Estrogens are a group of steroid sex hormones synthesised by neurons and astrocytes predominantly in females. In the hippocampus, estradiol plays a cruc
