Impact of varying maternal perigestational dietary folate intake on the development of cerebellar cortex in rat offspring

Philip Mwachaka, Paul Odula, Peter Gichangi, Adel Abdelmalek, Julius Ogeng'o · FENS Forum 2024

Background: Folate supplementation throughout pregnancy is recommended to support foetal and early postnatal brain development. The exact effect of folate on cerebellar development is unknown. This study looked at how maternal folate consumption impacts the structural development of the offspring's cerebellar cortex. Methods and materials: Adult (6-8 week old) female albino rats were randomly allocated to four groups: folate deficiency (folate 0 mg/kg), regular diet (2 mg/kg), folate enriched (8 mg/kg), and folate supra-enriched (40 mg/kg) premixed meals. The meals were started 14 days before m

Growth-factor receptor bound protein 10 (GRB10) is a signal adapter protein encoded by an imprinted gene, with roles in growth control, cellular proliferation, and insulin signalling. Grb10 has a unique, tissue-specific imprinted expression; the paternal copy being expressed in the CNS, whereas the maternal copy is expressed in most other adult tissues in the mouse and human. Nevertheless, although it is critical for normal behaviour in the mouse, the role of paternal Grb10 in brain growth has yet to be established. Here we investigate the brain-growth in mice lacking paternal Grb10 (Grb10+/p)
