Role of glycine receptors in the striatal network

Neha Prasanna, Bert Brône, Serge Schiffmann · FENS Forum 2024

The striatum is the largest nuclear complex of the basal ganglia (BG) associated with motor response where dopaminergic loops interact with local inhibitory GABAergic and cholinergic loops, facilitating or inhibiting simultaneously occurring neuronal signals. Whilst it was believed that the expression of Glycine receptors containing α2 (GlyRα2) subunit in the brain declines in adulthood, we have previously shown that besides GABA receptors, Medium Spiny Neurons (MSN), the projection neurons of the striatum, express functional chloride permeable GlyR, at adulthood, with α2 as the agonist bindin

Subcellular localization of GlyRα2 in spiny projection neurons

Yana Vella, Piccart Elisabeth, Serge N Schiffmann, Jean-Michel Rigo, Jelle Hendrix, Bert Brône · FENS Forum 2024

The glycine receptor alpha 2 (GlyRα2) is the only functionally expressed glycine receptor in adult striatal projection neurons (SPNs) and an important modulator of signal integration within the dorsal striatum. SPNs are a convergence point for glutamatergic and dopaminergic input, which is key to reward-motivated behavior. Thus, targeting GlyRα2 offers great potential to modulate striatum-orchestrated behavior. However, the impact of GlyRα2's subcellular localization on SPN activity regulation remains unclear.Neurons are morphologically complex cells, containing many subcellular compartments (
