Investigating the acute impact of sweeteners sucralose and Ace-K on ATP production and mitochondrial respiration in the hypothalamic GT1-7 cell line challenged with increased glucose

Aims. Acute consumption of sweeteners has become increasingly common in modern diets. However, emerging research suggests that the impact of sweeteners on the hypothalamus, involved in regulating food intake [CA1] and metabolism, may be deleterious. Several studies have raised concerns about their potential effects on brain cellular metabolism (Kohno D et al.,2016), suggesting a mismatch between detection of false sugar and the energy value actually detected. This study aimed to investigate the dose-dependent impact of sweeteners on these energetic processes. Methods. Using the hypothalamic ce

Omentin-1 in the mouse hypothalamic neuronal cells: Expression and effect on the regulation of gonadotropin-releasing hormone expression and secretion, viability, and apoptosis in GT1-7 cells

Omentin-1 (OMNT1), is adipokine involved in regulation of energy metabolism, insulin sensitivity, and reproduction. Our previous study documented expression and in vitro effect of OMNT1 on endocrine function of porcine anterior pituitary cells. However, its role in hypothalamus remains unknown. The present study examines expression and impact of OMNT1 on the mouse hypothalamic neuronal GT1-7 cells (in vitro model of GnRH-secreting neurons). The mRNA expression of OMNT1 and its potential receptor, insulin receptor, as well as co-localization of OMNT1 with GnRH, were determined using RT-qPCR and
