Functional connectivity of in-vitro neuronal spiking activity during rest and gameplay

Alon Loeffler, Forough Habibollahi, Moein Khajehnejad, Adeel Razi, Brett Kagan · FENS Forum 2024

In this study we explore network connectivity of in vitro neuronal systems of live biological cells within a real-time game environment of the game Pong. We use DishBrain - a system that embodies in vitro neural networks with in silico computation using a high-density multi-electrode array (MEA). We compare functional connectivity across resting-state sessions and gameplay sessions, during which the cells control a paddle able to hit a ball. The location of the paddle and ball are temporally and spatially encoded and presented to the neurons via electrical stimulation. We observe significant d

Investigating the development of the GABAergic system using human brain organoids

Chloe Hall, Sudena Wang, Michael K.E. Schäfer, Thomas Mittmann · FENS Forum 2024

Organoids derived from human stem cells present enormous potential to investigate aspects of human brain development. In this study, we hypothesised that brain organoids represent a suitable method for studying the shift in GABAergic transmission, from excitatory to inhibitory, during development. As several anaesthetics act via GABA receptors, we aimed to investigate how the anaesthetic propofol, known to be a GABA-A receptor agonist, may affect neuronal transmission in cerebral brain organoids at different developmental stages. Organoids cultivated from human embryonic stem cells (H1-ESC), w
