Cerebral venous blood flow regulates brain fluid homeostasis and neuro-immune surveillance

Marie-Renee El Kamouh, Myriam Spajer, Ruchith Singhabahu, Jean-Leon Thomas, Stéphanie Lenck · FENS Forum 2024

Lymphatic vessels and veins are closely associated in the dura mater around the venous sinuses, but how much venous outflow can influence the structure and function of dural lymphatics remains unknown. To investigate this vein-lymphatic interaction we developed a mouse model of cerebral venous outflow obstruction. Using postmortem and live imaging, we observed that glymphatic-lymphatic coupling is depending on venous drainage from the cranium and that lymphatic drainage impairment arises under conditions of venous drainage dysfunction. Long-lasting immune cell recruitment was also observed in

Stress promotes peripheral immune interactions at the brain endothelium

Johana Alvarez, Flurin Cathomas, Kenny L Chan, Lyonna F Parise, Aarthi Ramakrishnan, Molly Estill, Scott J Russo · FENS Forum 2024

Chronic psychosocial stress is a significant risk factor for the development of stress-associated psychiatric disorders, including major depressive disorder, which causes profound debilitation and has increasing worldwide prevalence. Pre-clinical and clinical studies have linked peripheral immune system alterations to stress-related disorders, such as elevated levels of proinflammatory immune cells and cytokines in circulation. Endothelial cells are critical components of the blood-brain barrier (BBB), as they interface directly with immune cells and their released factors, which can enter the
