Mapping of neuronal populations with light-sheet fluorescence microscopy

Giacomo Mazzamuto, Irene Costantini, Danila Di Meo et al. · FENS Forum 2024

Light-Sheet Fluorescence Microscopy (LSFM) is an excellent tool to study neuronal populations in large samples such as the whole mouse brain, thanks to its inherent three-dimensional nature and its high resolution down to the single cell. In this work we report on our latest technological developments in both hardware and software and their applications in several projects aiming at reconstructing the neuronal organization in both the human cortex and in whole mouse brain. Our latest LSFM setup is capable of simultaneous excitation and acquisition at four wavelengths, thus enabling fast reconst

A multimodal approach to investigate human brain mesoscale circuits with cellular and subcellular resolution

Irene Incerti, Giada Coluccio, Anna Maria Costa et al. · FENS Forum 2024

Progress in brain research is widely based on experimental approaches having the potential to advance the knowledge on fundamental biology. In particular, the increasing availability of data obtained by experimental techniques has boosted the development of data-driven mathematical models reproducing neuronal circuits. While cellular and subcellular resolution data have been extensively collected in animal models, experimental constraints have limited the acquisition of information about human neuronal cytoarchitectures and morphologies. In this context, high resolution data can be efficiently

Postnatal developmental dynamics of choline acetyltransferase (ChAT) and nerve growth factor (NGF) expression in rat oculomotor system

Diego Baena-López, Laura Morgenstern, Beatriz Betnítez-Temiño et al. · FENS Forum 2024

Choline Acetyltransferase (ChAT) serves as a reliable marker for motoneurons, including those within the oculomotor (OCM) system located in the brainstem. Although ChAT activity increases in spinal cord motoneurons during postnatal development, the evolving expression profile of this enzyme within the OCM system across various life stages remains unexplored. Previous investigations have indicated an elevation in nerve growth factor (NGF) expression in the spinal cord during postnatal development, mirroring ChAT patterns. Ocular motoneurons exhibit a distinct dependence on NGF due to the consti

Presence and characterization of neo-epitopes in mouse brain

Clara Romera Niño, Raquel Alsina, Marta Riba et al. · FENS Forum 2024

Brain ageing in mice leads to the progressive appearance of degenerative granular structures frequently referred to as “PAS granules” because of their positive staining with periodic acid-Schiff (PAS). It has been reported that these granules contain neo-epitopes that can be recognised by natural IgM auto-antibodies, and proposed that they intervene in a brain cleaning mechanism. Some preliminary studies seem to indicate that these neo-epitopes are also present in other structures than PAS granules, so the aim of this work has been to study and characterize the presence of neo-epitopes in mice
