The estrogen-immune axis: A key regulator of behavioural inflexibility

Mairead Sullivan, Sarita Dam, Angela Maria Ottomana, Martina Presta, Julia van Heck, Ilse Van de Vondervoort, Simone Macrì, Jeffrey C. Glennon · FENS Forum 2024

Aberrant insulin signalling was recently identified as a causative mechanism of Obsessive-Compulsive Disorder (OCD). Indeed, TALLYHO/JngJ mouse models of Type 2 diabetes are compulsive and anxious, with treatment with the Type 2 diabetes drug metformin generating partial rescue. Here, we aimed to establish the mechanisms behind this insulin-behavioural axis via proteomic analysis of brain and blood. Additionally, data from Genome-Wide Association studies (GWAS) of relevant disorders were compared using Ingenuity Pathway Analysis (IPA) (Qiagen). IPA analysis of proteomic results showed dysregul

Interleukin 1 signaling modulates pro- and anti-neurogenic effects through activation of interleukin 1 receptor type 1 on different cell types

Maria Smirnova, Marianne Monet, Daniel Nemeth, Henriette van Praag, Ning Quan · FENS Forum 2024

Adult hippocampal neurogenesis (AHN) occurs the dentate gyrus (DG) and is a dynamic process which functions in learning and memory. Increased AHN improves cognition and is therapeutic for neurodegeneration. However, loss of AHN is implicated in neurodegenerative disorders, thus understanding the mechanisms that modulate AHN could lead to targeted therapeutics. The master regulator of neuroinflammation, Interleukin-1 (IL-1), is a proinflammatory cytokine that signals via interleukin 1 receptor type 1 (IL-1R1) and decreases AHN. However, blockade of IL-1 signaling suppresses AHN indicating IL-1
