IL-1-PIP3 signal cascade mediates epilepsy and sleep disturbances

Shu-Hui Huang, Pei-Lu Yi, Fang-Chia Chang · FENS Forum 2024

Aims: Interleukin-1β (IL-1β) in the central nervous system initiates a PI3K signaling cascade, involving crucial membrane phospholipids phosphatidylinositol (4,5)-bisphosphate (PIP2) and phosphatidylinositol (3,4,5)-trisphosphate (PIP3). While PIP2's role in epileptogenesis is known, our study investigates the unclear role of PIP3 using PTZ-induced epileptic mice, exploring its impact on epileptogenesis and associated sleep disruptions. Methods: Our hypothesis proposed a pivotal role for PIP3 in the molecular mechanism of seizures. We intraperitoneal administered low dose of PTZ once every two

Monoamine signaling alterations trigger neuroinflammation and Alzheimer’s disease-like hippocampal deficits

Livia La Barbera, Annalisa Nobili, Paraskevi Krashia, Emma Cauzzi, Elena Spoleti, Maria Luisa De Paolis, Maria Teresa Viscomi, Marcello D'amelio · FENS Forum 2024

Recent preclinical and clinical observations identified alterations in the midbrain along the prodromal phase of Alzheimer’s disease (AD). These alterations include degeneration of monoamine neurons, resulting in deficits in ascending monoaminergic pathways in target areas related to AD, as the hippocampus. Incidentally, recent findings link the role of monoamines in the modulation of neuroinflammation. Here we sought to prove how the degeneration of the midbrain, leading to monoamine loss in the hippocampus, activates a neuroinflammatory response, in addition to alterations of synaptic plastic
