Amyloid beta 1-42 and alpha-synuclein proteins: Effects on transcription factor expression

Pelin Sordu, Merve Alaylıoğlu, Zuhal Yurttaş, Tugay Çamoğlu, Büşra Şengül-Yediel, Ebru Keskin, Duygu Gezen-Ak, Erdinç Dursun · FENS Forum 2024

Previous studies provided that Aβ1-42 peptide, a pathological component of Alzheimer's disease (AD), and α-synuclein, the major protein of Lewy bodies in Parkinson's disease (PD), may interact with transcription factors and affect gene transcription. Researchers have revealed that Aβ1-42 can be found in the nucleus, interact with DNA, and change the expression of many genes. It is known that α-synuclein functions as a transcription factor that binds to nuclear DNA and modulates the expression of many genes. Our study aims to investigate whether these peptides directly affect the expression of

Role of NCOR1 and NCOR2 transcriptional corepressors in myelin development and remyelination

Rubí Hernández Rojas, Ángeles Casillas-Bajo, Alerie Guzman de la Fuente, Jose A. Gómez-Sáchez, Hugo Cabedo · FENS Forum 2024

The transcriptional corepressors NCOR1 and NCOR2 are part of a multiprotein complex that downregulates Jun and triggers the transcription of myelin genes in cultured Schwann cells (SC) (Gomis-Coloma et al., 2018). Both corepressors are also expressed in oligodendrocyte lineage cells, both oligodendrocytes (OL), and in OL precursor cells (OPCs), suggesting that they might contribute to their development and function (Castelo-Branco et al., 2014). Using genetically modified mice we have previously shown that they are dispensable for myelin development in the peripheral nervous system (PNS). In t
