Photopharmacological control of GABAergic neurotransmission

Galyna Malieieva, Karin Rustler, Alexandre Gomila, Matteo Ranucci, Ulrich Zeilhofer, Piotr Bregestovski, Burkhard Konig, Pau Gorostiza · FENS Forum 2024

Inhibitory neurotransmission in the brain is mainly mediated by GABAARs. Disequilibrium in their function leads to many sever neurological disorders, such as epilepsy, anxiety, depression. Thus, development of allosteric modulators that would regulate the activity of these receptors with minimized side effects is of great importance. Photopharmacology is a unique tool for these purposes allowing precise spatial and temporal light-driven control of pharmacophores´ activity, and consequently of their target proteins.Pursuing the goal to develop a GABAARs positive photomodulator, we successfully

Role of Elkin1 in pain-sensing nerves

Jasmin Klich, Sampurna Chakrabarti, Gary R. Lewin · FENS Forum 2024

Mechanosensation is a process by which nerves encode physical stimuli such as cotton swab or pinprick into electrical signals that can be understood by the nervous system. But what is the molecular basis of mechanosensation, allowing this process to occur? New evidence suggests Elkin1 is an ion channel required for light touch sensation in mice and humans. Here, we explore its function in nociceptor physiology. Our results show that Elkin1-/- knockouts exhibit lowered mechanical sensitivity when tested in behavioral assays using a hindpaw stimulation. Electron microscopy showed no structural ch
