Metabotropic glutamate receptors and TRPC1 channel: Role in synaptic plasticity

Farah Issa, Xavier Yerna, Thibaud Parpaite, Nicolas Tajeddine, Philippe Gailly · FENS Forum 2024

Group I metabotropic glutamate receptors (mGluR) are involved in various forms of synaptic plasticity that underlie declarative memory by inducing long-term depression (mGluR-LTD). mGLUR-LTD results from an endocytosis of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs) underlying synaptic depression. The lab has previously shown that mGluR5 specifically activates TRPC1 channel, a Transient Receptor Potential channel that is highly expressed in CA1-3 regions of the hippocampus. To investigate the role of mGluR-TRPC1 in synaptic plasticity, and to decipher its downstre

Molecular mechanisms of remote fear memory extinction

Lisa Watt, Johannes Gräff · FENS Forum 2024

Elucidating the factors that facilitate or impede the extinction of remote memories is imperative for the advancement of targeted therapeutic interventions aimed at addressing maladaptive memory persistence. Despite the extensive body of research on memory processes, there is a notable paucity of studies focusing on the diminution of long-standing memory traces . Recently, our lab has identified a hippocampal remote recall engram that is a key player in remote memory attenuation (Khalaf et al., 2018, Science), yet its molecular underpinnings remain elusive. In this study, we seek to delineate

It is widely believed that synaptic transmission in the hippocampus plays a crucial role in formation of contextual memories. Data obtained in our laboratory implies that PSD-95 (postsynaptic density protein 95)-dependent synaptic plasticity in dCA1 area is required for updating of contextual fear memory. To check how inactivation of dCA1 area affects contextual fear memory extinction, we stereotactically injected the adenoviral vectors (AAVs) encoding DREADD receptor hM4 [AAV2.1:hSyn_hM4_mCherry]. Next we trained mice in contextual fear conditioning (CFC). We found that chemogenetic inhibition
