Role of PTEN in cortical stem cell lineage progression

Osvaldo Antonio Miranda Romero, Ximena Contreras, Florian Pauler, Mara Davaatseren, Nicole Amberg, Simon Hippenmeyer · FENS Forum 2024

The neocortex is an architecturally complex structure composed of a diverse population of neurons and glia. This well-defined structure and cellular diversity can be attributed to the tightly regulated and highly ordered cortical development program. PTEN mutations in humans typically result in severe macrocephaly, increased susceptibility to a variety of cancers, and Autism Spectrum Disorder (ASD). Here we explore the role of Pten in cortical development at single-cell resolution by utilizing Mosaic Analysis with Double Markers (MADM) to sparsely label and perturb Pten in our mouse model. We

Imprinted genes are expressed throughout the brain, exhibiting distinct allelic distribution in specific regions. To understand the imprinting status and their role in brain functions, we investigated the specific effects of uniparental disomy (UPD) by using a mouse model called Mosaic Analysis with Double Markers (MADM), which allows for the production of maternal and paternal UPD cell populations within the brain.By in silico enrichment analysis, chromosomes 2, 7, and 12 appear to be enriched in imprinted and behaviorally relevant genes, with different effects on post-natal development.
