Aims: Nuclear pore complex proteins (Nups) play critical roles in cell type-specific gene regulation by orchestrating lineage specification and maintenance of cellular state. However, their roles in adult brain remain largely elusive, especially in post-mitotic functional neurons. Accurate spatio-temporal regulation of neuronal gene programs is vital for neuronal plasticity and ultimately for learning and memory. We hypothesize that Nups play critical roles in neuronal plasticity and cognition through gene regulation. Methods: To address the importance of Nups for cognition and memory, we cond

Reduction of adult hippocampal neurogenesis is an early critical event in Alzheimer’s disease (AD), leading to progressive memory loss and cognitive decline. The nucleoporin Nup153, besides its role in nuclear transport, has been described as a key regulator of NSC plasticity through gene modulation. Here we investigated the potential role of Nup153 as target to improve neurogenesis in the 3xTg mouse model of AD in vitro and in vivo.We found that reduced Nup153 levels characterized NSCs from the 3xTg mice (AD-NSCs) and caused inefficient proliferation, migration and differentiation that were re
