Interleukin-33 as a player in axon remyelination in response to CNS and PNS injury

Małgorzata Zawadzka, Beata Kucharz, Katarzyna Konarzewska, Urszula Sławińska · FENS Forum 2024

Aims. Interleukin-33 is a protein enhancing Th2 cytokine production, initially described as an alarmin that is released from dying, necrotic cells to affect the immune response. Therefore, we aimed to examine the role of IL-33 in response to both CNS and PNS injury. Methods and Results. Using qPCR, Western blotting, and ELISA assay we showed significant up-regulation of IL-33 in two models of nervous system pathology in response to demyelination of the CNS white matter and sciatic nerve crush. We showed that IL-33 is produced exclusively by non-myelinating oligodendrocytes in the intact white

Myelin plasticity requires the expression in oligodendrocyte progenitors of NMDA receptors containing GluN3A subunits

Work over the last decade demonstrates that the myelin sheaths enwrapping axons, classically seen as static insulators, are highly dynamic and respond to axonal activity and experience. This phenomenon, called “myelin plasticity”, goes beyond postnatal stages into adulthood and plays a key role in remodeling neuronal circuits to support adaptive behavior or functional recovery upon injury. Yet the mechanisms are unknown. Here we show that NMDA receptors containing GluN3A subunits are expressed by oligodendrocyte progenitor cells (OPCs) and essentially required for activity-dependent myelin pla
