Chemotherapy alters the Kolmer cell of the choroid plexus

Parisa EmamiAref, Lucie Kubičková, Babak Bakhshinejad, Petr Dubový, Marek Joukal, Alemeh Zamani · FENS Forum 2024

Paclitaxel, a well-known anti-cancer drug, hinders cell division by stabilizing microtubules. Despite its effectiveness, paclitaxel causes neuropathic pain, possibly due to its impact on the immune system. The study investigated how paclitaxel affects CP macrophages, important for the blood-cerebrospinal fluid barrier. It focused on two types of Kolmer cell macrophages in a model of paclitaxel-induced neuropathic pain. Wistar rats received either paclitaxel or its vehicle (Ethanol and chromophore). Brains, including those from naïve rats, were collected at different intervals (1, 7, 14, and 21

Mechanisms underlying estrogen protection against paclitaxel-induced peripheral neuropathy: Modulation of upstream and downstream signals of high mobility group box 1

Chemotherapy-induced peripheral neuropathy (CIPN) is a dose-limiting adverse reactions in chemotherapy with cytotoxic anticancer agents. We have demonstrated that CIPN caused by paclitaxel involves high mobility group box 1 (HMGB1), a nuclear protein, derived from macrophages (Mφ) in laboratory animals (Neuropharmacology 141, 201, 2018), and that postmenopausal estrogen decline aggravates CIPN in patients undergoing paclitaxel-based chemotherapy (J Pharmacol Sci 146, 49, 2021). In this study, we analyzed estrogen modulation of upstream and downstream mechanisms of HMGB1. Female mice were subje
