Anterior cingulate cortex hyperexcitability in a mouse model of attention-deficit/hyperactivity disorder and pain comorbidity

Sandra Sanchez-Sarasua, Sarah Bou Sader Nehme, Marie Tuifua, Otmane Bouchatta, Marc Landry · FENS Forum 2024

Attention deficit/hyperactivity disorder (ADHD) is a common neurodevelopmental disorder in children and adults worldwide. It often coexists with anxiety, depression or alterations in sensitivity. Patients with ADHD report sensitization to pain and the prevalence of generalized pain is higher in ADHD patients (up to 80%) compared to control population (17%) (Stray LL et al., 2013). However, the mechanisms involved remain unknown. Neuroanatomical studies confirm that attentiveness and central mechanisms involved in pain transmission use identical neural networks (i.e., the anterior cingulate cor

Exposure to virtual nature decreases neural nociceptive processing and self-reported ratings during acute pain

Maximilian Steininger, Mathew P. White, Lukas Lengersdorff, Lei Zhang, Alexander J. Smalley, Simone Kühn, Claus Lamm · FENS Forum 2024

Engaging with nature provides numerous physical and mental health benefits, including reductions in self-reported pain. However, it remains unclear whether these reduced subjective ratings reflect modulations specific to pain, such as changes in low-level nociceptive or high-level cognitive-emotional processing or can be explained by domain-general factors unspecific to pain. To answer this question, we conducted a preregistered functional MRI experiment during which healthy participants (N = 49) received individually calibrated painful electrical shocks while exposed to audio-visual virtual n

Neuroinflammatory mechanisms of pain hypersensitization in a mouse model of ADHD

Sarah Bou Sader Nehme, Sandra Sanchez-Sarasua, Mairead Sullivan, Jeffrey Glennon, Eric Boué-Grabot, Walid Hleihel, Marc Landry · FENS Forum 2024

Attention-deficit/hyperactivity disorder (ADHD) is a common multifactorial neurodevelopmental disorder, with a prevalence of around 8% in children worldwide. It is characterized by symptoms of inattention, hyperactivity, and impulsivity and is usually associated with cognitive, emotional, and behavioral deficits. Clinical studies suggest that pain hypersensitivity develops in subjects with ADHD. However, the mechanisms and neural circuits involved in these interactions remain unknown. Our team has previously validated a mouse model of ADHD obtained by neonatal 6-hydroxydopamine (6-OHDA) intrac

Uncertainty in thermosensory expectations enhances an illusion of pain

Jesper Ehmsen, Niia Nikolova, Daniel Christensen, Leah Banellis, Malthe Brændholt, Arthur Courtin, Camilla Krænge, Alexandra Mitchell, Camila Deolindo, Christian Steenkjær, Melina Vejlø, Christoph Mathys, Micah Allen, Francesca Fardo · FENS Forum 2024

The human brain has a remarkable ability to learn and update its beliefs about the world. Here, we investigate how thermosensory learning shapes our subjective experience of innocuous temperature and the misperception of pain in response to harmless thermal stimuli. Through computational modeling, we demonstrate that the brain uses a probabilistic predictive coding scheme to update beliefs about temperature changes based on their uncertainty. We find that these expectations directly modulate the perception of pain in the thermal grill illusion as well as the constituent innocuous cold and warm
