Changes in mitochondrial respiration and mitochondrial dynamics induced by mitochondrial complex I inhibition in primary cortical neurons: Association with schizophrenia-like phenotype

Amanda Siena dos Santos, Luiz Felipe Souza e Silva, Vitória Cristina Araújo, Martina Raissa Ribeiro, Diana Zukas Andreotti, Larissa de Sá Lima, Ana Maria Orellana, Elisa Mitiko Kawamoto, Tatiana Rosado Rosenstock, Cristoforo Scavone · FENS Forum 2024

The goal of this work is to analyze the mitochondrial changes that Rotenone (Rot) treatment can induce in primary cortical neurons that could contribute to the schizophrenia-like phenotype animal model, previously demonstrated by our research group. For this, we previously determined the Rotenone (Rot) concentration in the cortex (1.325nM) from our animal model with a schizophrenia-like phenotype. Then, we treated the primary cortical neurons with Rot 1.325nM for 24h and investigated: the Rot cellular viability treatment by MTT and CytotoxGlo assay; CI enzymatic activity; oxygen consumption ra

The evaluation of PFCs (PFOA or PFHpA) on neural activity and survival in cortical neurons

Hyun Sung-Ae, Ko Moon Yi, Min Euijun, Minhan Ka · FENS Forum 2024

Perfluorinated compounds (PFCs) are organofluorine compounds containing carbon-fluorine and carbon-carbon bonds. They are used as water and oil repellants in clothes and pharmaceutical tablets, but have been found to cause neurotoxicity. However, it remains elusive whether PFCs affect neural survival or activity, or whether they alter different neuronal regulations in the brain. Additionally, there is a paucity of data on the regulatory mechanisms involved in PFCs-induced neural toxicity. In the current study, we report that PFOA or PFHpA differentially alter neural survival and activity in pri

Study on the efficacy and mechanism of ginsenoside in the prevention of cerebral ischemic stroke

Yan Pan, Shiqing  Zhang, Kaili Lin, Zhu Zhang, Ken KL Yung · FENS Forum 2024

First, on a cell model, primary cortical neurons were used to determine the safe dose of ginsenoside by performing toxicity assays with different concentration gradients of ginsenoside using MTT assay; Then pharmacodynamic screening was performed using primary cortical neuron oxygen-glucose deprivation reperfusion (OGD/R), and primary cortical neurons cultured to day 7 were replaced with sugar-free DMEM after drug preadministration for 2h.Then the neurons were placed in a hypoxic chamber containing 95% N2 and 5% CO2 for 4 h, and after that they were switched to the normal medium containing gin
