This EMBO–EMBL symposium connects the molecular machinery of DNA replication with genome stability and human disease. It examines replisome structure and dynamics, control of origin firing across tissues, chromatin and epigenome maintenance, chromosome organisation, replication-fork protection and repair, and the consequences of replication stress for development, ageing and cancer. The programme links mechanistic advances to cancer biology and possible therapeutic approaches. All programme times are Europe/Berlin. Opening remarks on 20 October begin at 13:30, followed by the keynote at 13:45

Modelling Koolen-de Vries syndrome in neural organoids

Brooke Latour, Spencer Shute, Iris Teunnissen van Manen et al. · FENS Forum 2024

Koolen-de Vries syndrome (KdVS), also known as 17q21.31 microdeletion syndrome, is a rare neurodevelopmental disorder characterized by developmental delay, intellectual disability, and congenital malformations in multiple organ systems, including the brain. KdVS is caused by haploinsufficiency of KANSL1, a gene encoding a protein involved in chromatin remodeling during interphase and microtubule stabilisation during mitosis. KANSL1 is widely expressed in early development, but its role in early brain formation is not characterized. Using patient and genetically-edited induced pluripotent stem
