Modelling regional specification in brain organoids using a novel mesofluidic device

Soraya Scuderi, Alexandre Jourdon, Tae-Yun Kang, Liang Yang, Alex Nelson, Feinan Wu, George M. Anderson, Jessica Mariani, Livia Tomasini, Alexej Abyzov, Andre Levchenko, Flora Vaccarino · FENS Forum 2024

Morphogen gradients play a critical role in directing cell fate specification throughout development but are not yet implemented by current in vitro models. Human induced pluripotent stem cell derived brain organoids recapitulate early cellular and molecular mechanisms of brain development. Because they lack spatial organization to fully mimic the in vivo topographical organization, we introduce a new method to model early neural tube patterning by exposing organoids to continuous gradients within a five-chamber mesofluidic device. We hypothesized that the introduction of oriented signaling ce

Recording cellular memory to unveil the mechanism of brain memory

Yumiko Kawamura, Valentina Khalil, Taro Kitazawa · FENS Forum 2024

Cell identity and states are dictated by gene expression programs, influenced by a range of epigenetic modalities. In the neuronal system, these mechanisms underlie various functions, such as maturation and memory formation, and their deficits are linked to various neurological disorders. Recent advancement of single-cell sequencing technologies, including scRNA-seq, has accelerated our understanding of the cellular diversity with respect to molecular profiles. However, existing high-throughput sequencing methods have an inherent limitation that they can primarily provide a static ‘snapshot’ o

Spinal cord injury (SCI) is a complex disorder that triggers a cascade of biochemical processes interwoven by cell-type specific responses after the initial injury. This progressive cascade is collectively termed the "secondary injury", and involves inflammatory cell infiltration and cytokine release, apoptosis, demyelination, excitotoxicity, ischemia, and the formation of a fibrotic scar with an astrocyte border. Here, we turn to single-cell technology to unravel the underlying cellular and molecular tapestry of SCI. We compiled a series of comprehensive single-cell atlases of the injured mou

Unraveling epileptogenesis mechanisms via scRNA-seq in Scn2a p.A263V mouse model

Xian Xin, Malte Stockebrand, Birgit Engeland, Dirk Isbrandt, Konstantin Khodosevich · FENS Forum 2024

Our study investigates mechanisms of epileptogenesis caused by the p.A263V mutation in the SCN2A gene using a Scn2a p.A263V mouse model. The SCN2A gene is known for its role in neurodevelopmental disorders with epileptic presentations. Our objective is to understand the molecular foundations of neonatal neuronal hyperexcitability and its evolution into adult network disruptions, employing scRNA-seq. Neonatal homozygous (hom) mice showed seizures starting around P3, with heterozygous (het) mice also exhibiting early seizures. Despite recovery in het mice, persistent network alterations were obs
