CYFIP2 Arg87Cys mutation: Unraveling its role in early infantile epileptic encephalopathy

Elona Ndoj, Luca La Via, Matteo Bertoli, Alessandro Barbon · FENS Forum 2024

CYFIP2, a component of the WAVE regulatory complex, plays a crucial role in regulating actin dynamics and synaptic function. Through genetic analysis of patients diagnosed with Early Infantile Epileptic Encephalopathy (EIEE), novel mutations in the CYFIP2 gene were identified. Functional studies revealed altered synaptic plasticity and neuronal excitability associated with these mutations. Furthermore, behavioral assays in animal models harboring CYFIP2 mutations demonstrated phenotypes consistent with epileptic encephalopathy. These findings underscore the significance of CYFIP2 in neuronal d

The interplay between mTORC2 and oxidative stress in neurotoxic models of neurodegeneration

Marija Jeremic, Andjelka Isakovic, Vladimir Trajkovic, Ivanka Markovic · FENS Forum 2024

1-Methyl-4-phenyl-pyridinium (MPP+) and 6-hydroxydopamine (6-OHDA) are the predominant neurotoxic agents used in the research of Parkinson's disease (PD). The mTORC2 is a significant signaling pathway whose specific functions and mechanisms remain partially elusive, especially in the context of PD-related oxidative stress. We aimed to determine the interconnection between mTORC2 and oxidative stress in MPP+ and 6-OHDA neurotoxic models of PD. All experiments were performed on the human neuroblastoma cell line, SH-SY5Y. Cells were divided into two groups: control (consists of control cells and

Silent killers in your everyday products: Unveiling the neurotoxic effects of endocrine disrupting molecules

Camilla Corrieri, Agnese Graziosi, Luca Ghelli, Giulia Sita, Patrizia Hrelia, Fabiana Morroni · FENS Forum 2024

Endocrine disruptors (EDs) are ubiquitous compounds with the ability to interfere with endogenous hormones. Molecules identified as EDs are highly heterogeneous, but most of them are man-made chemicals, such as ethinyl estradiol (EE), medication, perfluorooctanesulfonic-acid (PFOS), surfactant, and diethyl-phthalate (DP), plasticizer. The constant exposure of the global population to these molecules can lead to an induction of toxic effects, especially on the central nervous system. Among the different mechanisms by which EDs could influence human health, of interest is the modulation of micro

Study of lipid droplets in brain aging and neurodegeneration

Emanuela Grillo, Elisabetta Pingitore, Luca Tirinato, Yuan Li, Lene Juel Rasmussen, Laura Berliocchi · FENS Forum 2024

This research project investigates the role of lipid droplets (LDs) in neuronal survival and dysfunction, focussing on two human in vitro models of neurodegenerative diseases (NDs): neuroblastoma SH-SY5Y cell line exposed to 6-OHDA for Parkinson’s, and primary human fibroblasts from Alzheimer’s disease (AD) patients.Valid and reliable experimental tools for LDs detection and quantification were set up in SH-SY5Y. Confocal laser scanning microscopy was combined with image analysis, allowing an automatic and objective quantification of LDs in basal conditions, and in response to LDs modulation a
